Male playing as both. I usually take multiple characters through games and generally take a 50/50 mix of genders. I do this to explore as much of what the game has to offer as I can.

I am old enough to remember the days where playing as a female was an exception to the rule so I haven't really gotten over the "oh, wow, I can play a woman" mindset when I get new games. I was a huge fan of Duke Nukem so I live vicariously through Mass Effect with my bleached blonde, black lipstick wearing (cause there's no 5 o'clock shadow available, y'know), gun toting, trash-talking female Shephard. I play male characters if there's a story that goes with it (such as a romance being only available to specific genders).

Female playing a female. I always prefer games where I have a choice to play as a female. Plus to me I feel odd and bulky running around as a guy.

Female playing as female to start. Probably one of my characters from the last time around, actually. I'm more familiar with them, and Jennifer Hale as Shep. (People say Mark Meer's taken a level in Voiceacting. I'd love to see it before I try.)

Two people, one account, so both and both here. For us, the other watches while the other plays, mostly storyline and romance scenes and whatnot. It's a lot easier than having to create another file to play through for the achievements.

I like to make my RP characters based off of me, so I usualy choose male, model them after me, and give them my name.
For female or evil characters, I name and model them off of my niece.

I'm male, and playing female. I don't know really why, but somehow playing female has always been more interesting somehow. Perhaps it's the curiosity to see what's it like on the other side of the fence. On some games the female animations can be better than male counterparts, but that's not an issue in Mass Effect. Perhaps it's just the fact that Shepard is more pleasing to look at if she's a she. Also, a female special ops character is more unorthodox and therefore more fun to play.
And of course then there's voice actor vs. voice actor, and I prefer Jennifer Hale. I'm something of a fan of hers
Even more so when I found out that she played both Deionarra and Fall-From-Grace in Planescape: Torment, my favourite game.

Male. Almost always play female characters, because the guys in video games tend (with some exceptions) to look like bulky body builders. It just isn't appealing, especially when I'm playing a caster class.
Femshep also has a better voice actor.

I'm a female & I'll be playing my FemShep. Considering that I usually have to play a guy in most games, when I can actually play a female I prefer to do so. However, in the interests of achievements I might make a male character sometime.

My first playthrough of ME2 will be as my male Shepard (blame Miranda for that), but as with ME1, the majority of my plays will be with my female Shepard.
Why? Because I like diversity. Ninety-nine percent of the games I play or own star a male as a main character. I like to mix things up. Jennifer Hale being an awesome VA also helps, too. More games need to star female characters, or at least provide the option to make female characters to star in the game. The more diverse things are, the better they are.

I couldn't choose so I ended up with 2 canon Sheps. As a male I can relate more to a male shep, but then there is Jennifer Hale...

One reason is I don't like how the standards look and I can at least make a Male Shep who looks half human, my attempt at a custom Fem Shep resulted in something that looked like the love child of Joan Rivers and the Sarlac.

I am male and I play both, I have a lvl 60 male and female, I tend to make more females though. I am not happy so far with the femshep romance options, so my femshep will probably not be cheating. Male shep is going to have a rough time though, good thing I have a run through with no LI.

My most complete save file is a femshep. Otherwise it would be my male infiltrator.
