If you're using the stuff that came with your PC, then I'm betting it's just adequate for getting internets done.
Once you get used to the keyboard / mouse combo you'll find it hard to go back to the controller.

Getting used to not being able to 'quickscope' using sniper rifles and the likes also takes some time to get used to.

No auto-aim on PC, so you'll be missiling fewer swarmers and scoring more head shots.
You can also learn to "run the pizza" - a very helpful skill -and iirc, only PC players can do this.
Charging weapons (e.g., Acolyte, Geth shotgun) is hold right mouse key.
Spend a evening playing some bronze matches just for fun, and you'll get the hang of it in no time.

Yay for Carpal Tunnel! This time I get to include all of my fingers and not just the right trigger finger.
Alright, I'll give her a whirl and see if i don't end up as a decorative rug for the nice marauders and such to wipe their feet on

Practicing is the best way to find what is not comfortable and correct it. Each feeling is different.

My main mappings are:
RMB: shoot
LMB: scope
MMB: Power 1 (e.g. throw)
Wheel forward:Power 2 (e.g. dark channel)
Left shift: Medi gel (for reload cancelling)
But as guigui says - everybody are different , try out a few things.
To make up for the lack of sixasis, enjoy:
Not waiting around staring at load screens
Headshotting collectors
Getting through more games without disconnects
Looking around whilst sprinting.
Reload cancelling

well that's not very helpful, I know plenty of Novagods that are fully Gold capable. I already understand how the game itself works. My current problem is transitioning to the PC from PS3. Seeing as I don't play very many PC games yet, I was requesting assistance in becoming quickly comfortable with PC controls. Quite a few forumites have made helpful suggestions (such as remapping, upgrading equipment, etc.) and have described what it took them to transfer from console to PC. Most of all, I have realized the value that simple practice is going to hold for becoming comfortable on PC thanks to these wonderful contributors.

Right handed mouse
- Arrow keys for movement
- Enter (Run, cover, jump over cover)
- Num Pad 1 (power 1)
- Right shift (power 2)
- Right control (power 3)
- Num Pad 0 (Medi gel)

- Num Pad 7 (Ammo pack)
