Assuming that you are playing on a computer, as long as your copy of ME3 remains linked to your Origin account (I see no reason why that would not be the case), you should be able to install the game on any machine you see fit.

No, you can download the game through origin even if you bought a hard copy of the game.

I don't know if I need to get a new disk or can install my old one again, and in case I go with a new one would I keep my manifest and challenges? This is bothering me a lot since I could not face to begin from zero one more time, really

All your multiplayer achievements and unlocks are linked to your origin account. Provided the servers are still open, you can take a 6 month break and come back. You're stuff will still be there.

No! You definitely don't need to buy a new disc/copy of the game.

Just install Origin and log in with your credentials. (If you do so via the DVD then you have to go through the update process.)
In the main Origin window "My Games" section, all your games related to your account should show up. From there you can download and install them.

-> right click on the game -> Show Game Details.

Make sure you install ALL the Multiplayer-DLCs.
