I don't know if it's Shep's response or merely the way she says it (or possibly a combination of the two) but I always get a good chuckle every time I hear it.

"Virtual ass into actual dust" always makes me LOL.  The fact that the stupid VI told him I was there... not so much.

Oh instant for me, and a inside joke with all my friends. When your in a firefight and the AI enemies keep screaming "Enemies everywhere!!" and then when your squad dies and its just you the continue to scream "ENEMIES EVERYWHERE!!!", hands down my favorite,cracks me up

I don't remember the exact quote, but its the one where you do the rachni mission on the freighter and Wrex makes a comment about the irony of him destroying the rachni again. Also any of Wrex's comments while on the elevators.
