I think I have many favourites. But the highlights for ME1 are:

Ashley: A million lightyears from where humanity began, and we walk into a bar filled with drunk men drooling over half-naked women shaking their asses on a stage. I can't decide if that's funny or sad.
Kaidan: What, you don't think they are here because of the food?

Ashley (on Noveria): Nobody lick any poles.

Ashley: Why is it when someone says "with all due respect", they really mean "kiss my ass"?

Kaidan: Oh my god! (<-- somehow when Kaidan says it, it cracks me up)

Kaidan (renegade): Fifteen thousand years to figure this out, and it's down to twenty-five mutiniers. Way to go Team Milky Way. (sarcasm = win! )

ManShep (modded M/M romance): Kaidan I don't believe I had the pleasure of you serving under me. Don't you think it's time to rectify that? (ManShep says it so emotionlessly, but it does crack me up badly)

I absolutely love that pause in the middle of his explanation, as if Garrus never fully thought about the profit in selling Krogan testicles until that point.

Whenever I get to that conversation I turn the volume way down for crew members, and all the way up with full bass when Sovereign speaks. You feel small and irrelevant already, and the implication there are more of them than you could count in your lifetime furthers the feeling. For that moment, you're not fighting a war; it feels like the universe has bent its fundamental laws to come get you. It may not have the cachet of other lines, but it works for me.

Other winners include "And I represent the second-guessing of galactic law" (even my Paragons give him this one, it has such living-the-movie punch along with "I can bludgeon pretty hard"), "No we don't, sir" (the moment my Earthborn/War Hero 'gang kid done good' decided she liked the hell out of Ashley), and "Well, if you're going to make a krogan statue, might as well make it big, right?" (the only thing to come out of Kaiden I ever liked - a Shepard for every class at level 60, and he's never made it off Virmire).

EDIT: And one I've never heard before, since I just ran past Yaroslev to talk to the Hot Labs Mira...with Liara in tow.

"How you tolerate literal-minded VIs is beyond me."

FURTHER EDIT: And of course, the Galactic Hypocrisy Award acceptance speech the turian Councillor offers.

I thought the entire exchange with Sovereign was AMAZING!  Best of the game.  I say it is my favorite quote of any game I have ever played.  I still get goose bumps when I talk about it with other gamers.

I cannot believe I forgot this one all this time. That's what comes of not checking the Codex much after your first playthrough. Laughed for a good five minutes the first time.

Until the invention of gunpowder weapons, 'eaten by predators' was still the number one cause of krogan fatalities. Afterwards, it was 'death by gunshot'.

EDIT: Somehow, I always find at least two every time I sit down to play again, though I guess it's not technically a quote. However, the eyebrow Shepard raises at Stirling (almost a "<_<" expression) on the Noveria docks in response to "We're the law here, show some respect!" is incredibly eloquent for a wordless gesture.

Seems like you haven't played ME1 assignments all that much? You'll often fight in bunkers filled with enemies who would more often than not spam above mentioned one-liners non-stop during combat.

It's not really a quote but I find it to be pretty hilarious.
