Oh! If we're imagining straight-to-future events :D.... (and ignoring Mac's recent words hehehe lalalalalala):

-Yahg or Krogans challenging the galaxy to varied levels
-Geth or Virtual Aliens (on their own platforms) challenging the galaxy to varied levels
-Quarians or Raloi engaged with the galaxy to varied levels
-Rachni or Kiriks challenging the galaxy to varied levels
-The above three factors are not central to main storyline, but ways to show some impact from choices (in individual missions, not arcs) and introduce new races/societies to the mix for future exposure in the franchise

-I won't address S/C/D options much due to anything being possible. Heck, even on the literal bent, those green squiggly lines in Synthesis could very well fade into the galaxy (all material) within weeks, so heh. Again, anything is possible.

-Salarian Civil War! Those agreeing to the imprinting to Dalatrasses (which isn't something previously focused on in Mass Effect), and those working with or lead by STG in rebellion. Exposure of Salarian rulers secretly being closer to Cerberus-lite (though many Salarians will not stand by this, and will join in rebellion).

-Asari exposures and downfall. How much did they know? I can see Asari being as disgraced as it seems the Consort was (though yeah yeah, I suppose that was due to a glitch?). I would love for them to do some soul-searching and explore their original roots more. I'm not against the species at all; I only think that some hubris might be in store and it may be illuminating.

-Humans as greatly spread throughout the galaxy. Not like Quarians really, but more as wide society that influences everything, especially due to Shepard's actions. Think of how it seems other species were considered 'N7' in co-op, and take that idea to an interesting conclusion :). We may see a galaxy without a centralized rule anymore, but instead with many factions and characters attempting to 'make sense of it all'. Earth may be rebuilt (if we are to take the Extended Cuts slides as the full truth), but I can see Humanity as changing their plan from 'colonization push' to 'spread and assist everywhere'. They may be as welcome, or at least allowed, everywhere just as Spectres would be - at least generally speaking.

-I can see Bioware taking a step back from the War-pew-pew and making things a bit more Star Trek (especially TNG). They very well may have a *ton* of action happening, but the story may involve a lot more exploration, investigation, and the occasional diplomacy. ME1-3 setting the foundation, but 'ME4+' using newer tech (both in RL consoles and in-universe advancements) to let us get out of our 'Reaper tech' bubble and more easily understand the galaxy at large, and maybe beyond the Milky Way.

-That said, co-op and even versus modes are almost givens, in some form. There are ways to make this work (with maybe things like Salarian Civil War being a hypothetical canon event, no matter what we do in ME3).

-Reapers still exist. Or rather, one-off stories involving the Reapers and/or their legacy could exist as tie-ins to the old world, but they wouldn't be core to anything. This is at least one view I have on this. At least.. I don't expect people to go "Oh Reapers? What are those? Oh yeah, we fought those once." and ignore it entirely otherwise. lol

-Female Krogan fully shown
-Female Salarians elaborated upon
-Female Turians elaborated upon
-Male Quarians fully shown
-Possible retcon-ish of Quarians to make it a mix of that bad Photoshop and some of the better fan/in-studio images of them
-Improved Human models, closer to the FMVs we see in ME2-ME3 promotions


Possible topics I wouldn't mind explored, as long as they're VERY WELL DONE:
-time travel (the iffiest one)
-organic evolution, where it really leads to, what it means
-synthetic evolution, where it really leads to, what it means (aka a world without the Cycle of Reaping imposed)
-exploring what those weird things Prothians and Asari and such do. Entering/experiencing memories?
-virtual reality
-facing the unknown, in every way, like starting ME1 all over again
-starting to take things to the universal scale, but I'd prefer it to still stay within the Milky Way for the most part

All in all, I want to explore the Mass Effect universe to the greatest extent possible, now that the imminent threat of the Reapers seems to be gone. At the same time, I want a new arc, new revelations, and at least the START of a bigger threat or goal to approach us all. Some may be strongly against this, but I'm only saying it should be the START.

-We don't need to be the Alliance Soldier in all of this. A different take on things is welcomed.
-We don't need it to be about RAWR GIANT ROBOTS for there to be an even larger threat. Imagination is valued here.
-Don't get SO attached to plot and characters. Sometimes, the experience itself can transcend that, and plot and characters only facilitate a great overall story.

Species I'd like to be on my ship:
-If we take premise that I said at the beginning (somehow a direct-future sequel lol), then I think we may be done with Quarians, Krogan, Geth joining as party members......
-HOWEVER, there may still always be a PLACE to have Yahg, Raloi, and more join. I would like that a lot.
-Turians/Asari/Salarians I'll always welcome. Especially another Salarian.
-Better human party members. ME1-3 was hit and miss with how they created human party members imo.
-I honestly still want a very interesting Batarian :S

Some days, like today, I want to have a prequel with mostly action, little story and barely any choices just so I can witness BSN explode and rain down millions of salty, bitter tears.

Other days, I want to be a space pirate. Grand Theft Spaceship: Mass Effect. Please, Bioware, make it happen! I want to be able to switch between characters on the fly. I want epic, fully controllable high speed chases. I want to be able to purchase, trade, upgrade and personalize ships. I want to explore and steal things. I want to deal red sand. I want choices. I want dialogue that isn't like an interview , but remember what I said about choices. More clothing options. More armor upgrading and personalization. Moar weapons. And powers. And zero G environments, rain, snow, wind, earthquakes, volcanoes...explosions.

I want a more personal journey. Something not so high level. Something (dare I say) not so significant on the galactic scale, however very significant to the characters involved. I want the next game to once again make me feel like I am a very very small speck in a massive sea of a galaxy. I want to feel the wonder I felt in the first game. Hopefully its a stand alone game. Self contained story with no commitment of a trilogy. This way the writers don't have the burden of worrying about continuing a characters story across multiple games in order to deliver fan service. Giving them less constraints and more freedom to kill off whoever and tell the best and most dramatic story they can.

I don't want:

The next villain trying to live up to the Reapers
The next protagonist trying to live up to Shepard
To see any of Shepards crew
To have another linear corridor shooter (get back to an exploration type RPG)

Small scale. Focus on a single system and the politics of surrounding systems and relations with larger organizations and government. Flesh out the planets and give a wide range of areas on the main planets to explore.

Also must be AU where only the First and True Mass Effect happened.

More than one person beat me to it, but yeah. I want to be a leader of a merry band of outcasts on a small freighter, taking jobs to keep the operation going.

It would be my favourite scenario set in my favourite universe. (No it's not done-to-death, it's a classic.)

This is pretty much what I want.
I wouldn't mind a little grandtheft spaceship either, a story thats more about your character and friends than the future of the entire galaxy. 
Some choices can ofcourse have impact on a lot of people but I wouldn't want it to mean the death of an entire species or the end of galactic civilization.

I think a story set about a year after Shanxi would be interesting, either from the human perspective of having just experienced the shock of discovering other species and having fought one of them, or from an alien perspective of dealing with these newcomers from the Sol system.

Not simply fought one of them - but got crap lobbed at their colony from space. No humans had fought anything like that. The shock of that would linger for awhile. Just seeing stuff like this would explain why many humans fell into xenophobia and Cerberus was created.

I always thought it was a copout after ME1, when we couldn't roleplay lingering resentment against Turians much. Garrus became too much of a "bro", and shaped Turian relations (not that that's a bad thing either, but it oversimplifies things).

Although I liked the ME-trilogy dispite its inconsistencies, flaws and over-the-top antagonists (the galaxy-scale terror the reapers represented), I would like something on a more profane level (not the classic overwhelming evil vs. the small forces of good). I'd like it to be a bit darker and more naturalistic. Something that is less epic and more intricate:

More mistery, less black and white characters and decisions, more "human" element, a bit more intimate of a story, more charismatic antagonist with more fleshed out goals and character (which you can relate to on a level), the same for the protagonist we play, more politics (galactic lore) and less of a high-fantasy feel. 

I think Karpyshyn and Walters did a good enough job (yes, they did, even if they are not the best of the best, which is obvious), but there's so much more potential in Casey Hudson's universe than it was showcased in the trilogy. 

The ME-trilog was a nice learning ground for those who are involved in the creation of the ME universe and it was a relatively successful experiment, but I think now that many lessons were learned, it's time to bring it to another level. I'm hoping for something much better than the ME-trilogy (which I like very much as a game).

It would be also good if it wasn't aiming for a cinematic block-buster kind of feel. Yeah, there should be epic and spectacular moments, but they should not steal the game from the story-telling. ME3's greatest mistake was that it tried to be way too epic. It was epic as f.ck, but sometimes it felt a bit too much. Sometimes less is more.

I'd like it if the Batarians would play a bigger role, maybe with a squadmate. They have so much history with the Alliance, being their main opposition in the Galaxy. (I really don't understand why they received so little attention during the trilogy). They also represent so many things we dislike but which are present in human history or global community (Slavery, Terrorism, Isolationism, Banditism). It's like an ugly mirror image of humanity. On the other hand new aliens who aren't just humans who look different would be interesting.
I'd also like more planetary exploration, with a ground vehicle like the mako but with better controls.
And I want less auto dialogue, less forced perspectives and more room for roleplaying different characters.
