Several companions can be 'hardened', or changed, through your choices in a quest (the affected companions side-quest). The hardening has some major storyline consequences, mostly applicable to the end game. It affects the characters' dialogues, and the overall attitudes of your party.

It means you're making them less naive/childish about certain things. You essentially tell them "buddy you gotta start looking out for yourself because the world is not a nice place." You should read the wikia page: Hardened for the specific scenes/dialogues where Alistair or Leliana being hardened or not makes a difference. (spoiler alert, of course)
Personally I always harden them both. A determined Leliana makes more sense with what she ends up doing in DA2/Asunder, and an assertive Alistair makes the Landsmeet and its fallout much more interesting.
