You can make him jealous, sort of. If you have started a romance with another person before the chess scene and you choose to flirt with Cullen saying 'we should spend more time together', he would call you out: '(not exact wordings) Don't you prefer the company of XX' And you can respond him by friend-zoning him
or the-I-forget-what-it-is option.
And of course, if you are romancing others and you ask him to talk alone, he will say something like 'I thought you are...involved with someone else.'
Ah, not one for drama, our Cullen? Good for him, I guess.
It actually fits, as I can't really imagine him bothering with stuff like that.
If you're in a relationship with Cullen, he won't have any response to you flirting with others. In fact, I don't think any of the LIs have the type of jealous reactions we've seen in other games (I'm far from an expert on all of them, though). The only thing that seems to occur is other people sometimes turning down your advances once you're in a relationship, as Raccoon mentioned.
If you're playing a female, feel free to flirt with Dorian and/or Cassandra at every opportunity. They love the attention.

Yeah, the flirts you get when the relationship is locked out are pretty tame, so it would be kind of weird if any of the LI's were to get jealous. IIRC in Origins it would be a more forward (or maybe not? can't remember), I believe you could actually cheat. I was way too much of a pansy to try it.
I remember playing Origins and thinking that Zev was the best friend ever until Alistair had a fit and demanded I choose between them.
