I tend to keep the "original" party a lot when I play games like these. At least, the first playthrough.
Cassandra, Varric & I don't know about the mage. For now, let's say Vivienne.
I don't usually stick to one particular party, so there will probably be a lot of switching in my first run to get a feel for different companions' abilities and personality. I do know that I'll play a rogue first time out, and I suspect Varric will get a lot of time in the field because I'm used to him being a fairly constant presence in DA2.

I tend to switch around a lot in my first playthrough so that I can get a better understanding of who these people are and how they react to certain things. Then in subsequent playthroughs I'll just choose whoever fits my Quizzy best. Varric will probably be around a lot the first time, though, because I'm familiar with him.
I can't really determine the "main" party until I see how the party dynamic works with the story. In DA:O, Alistair was my solid right hand for the entirety of the game, due in large part because his personality was my favorite, but also because he had the best reason to be among the entire cast of companions, with Leliana (archer) and Morrigan/Wynne (both arcane warrior) as my regulars. In DA2, it was Varric, Isabela and Merrill as my solid team throughout most of the game, alternating with Aveline for the "official" quests in the game. DA2 kind of benefited from the team not really be involved in a main quest together like Origins, so I had no trouble completely ignoring some companions for long periods of time without feeling like they're lingering at camp somewhere.
While I wont be using them for every mission as I like to try and use everyone, my "main," party make-up will probably look something like this for my first three playtroughs -

DHMG (Because of his dramatic hands and his mustache, and would honestly love to see my Dalish travel with a magister (hopefully)
1st playthrough

I'll use the other on following playthroughs.
I won't do such a thing when I don't know my companions (accept Varric). I'll rotate them.
Depends on my Inquisitor. Each character I roleplay would travel with different people, I think.

I got no clue really.
But since there are nine companions, I will play 3 times to get each of them a full game for sure.
Depends though on the combat set up.
If I want to I will just play on casual and make a full party from one class.
