Save Game Importing
The Dragon Age Keep is a web-interface that will allow you to create your own world state by making all the decisions for everything from Dragon Age:Origins through Dragon Age 2: Mark of the Assassin, and everything in between. This will be tied in to your Bioware Account, so no matter what system you play on, you can import the saves.
It is unknown at this point if you will be able to actually import from the old games, or if you will have to create them from scratch within the Keep.

Locations
We will be travelling through Orlais (including the Dales) and Ferelden.
The visitable areas will be much larger, encouraging a sense of exploration with a sort-of "open world" feel. (Although it's not one big continuous map, such as Skyrim).
One area, The Hinterlands, is described as being a "medium-sized" area. And it is larger than the ENTIRETY of Dragon Age: Origins combined![5]

Dialogue System
There are three possible "wheels" that you can encounter during the game:

The Tone Wheel is the "standard" option that you see most often in DA2. You are selected dialogue for the purpose of conversation and exposition. Your character's choices reflect his overall attitude and demeanor. The tones from DA2 (Diplomatic, Sarcastic, and Aggressive) have been replaced with Noble, Clever, and Direct.
The Action Wheel is for making a hard-line choice between two (or more) options. It is not tone-based (such as the Accept/Deny quest options in DA2), and all options are "neutral" in terms of tone. To avoid confusion, each option will tell you explicitly the intent of the selected option.
The Reaction Wheel is for reacting emotionally to a situation. Instead of choosing your tone, you choose an emotional reaction (such as "Sad", "Enraged", "Confused", etc.). The "Stoic" option is always available for a reaction-neutral response.
Conversation will also be more dynamic, allowing you to just walk away mid-conversation.[5]

There will be SOME form of online or multiplayer aspect to the game. Beyond that, though, no details have been announced.
It could be full-on multiplayer, or it could just be uploading selfies of your Inquisitor to the Forums. No clue yet.

You can choose your gender as always, and can choose between Human (Noble if not a mage), Dalish Elf, Surface Dwarf, and Vashoth Qunari.
There will be two voices for each gender (one American, one British) for you to choose from when creating your character.

The Inquisitor is NOT the so-called "Old God Baby" from Dragon Age: Origins.

Combat
Combat will be far more tactical, placing emphasis on elements such as positioning, elevation, chokepoints, and preparation, with a strong focus on teamwork.
There will be no level scaling. This means that you may encounter areas or enemies that are too difficult, and you'll have to come back to them when you are stronger.[5]
The standard difficulty options will be back as before (Casual/Normal/Hard/Nightmare), but Normal will be slightly more challenging than before.

Romance & Companion Interaction (General Information)
Romance will be in the game, with no stronger or lighter a presence than it has had before.
It is unknown at this point how the potential Friendship/Loyalty system will work. Only that it will NOT be the same as either Origins or DA2.
There will be more romance options available than in any previous Dragon Age game to date.[5] Dragon Age 2 had 5, so this implies at least 6 romance options.
(There was confusion on this point, in that it was initially stated to be more than any "previous
Bioware game". But this has been clairifed in
Mike Laidlaw's twitter to be "previous
Dragon Age game".)

Weapons and Armor
You will be able to change your companions armor and weapons (even weapon types), although they will maintain a sort of "personal style" to anything they wear. [4]
There will be a crafting system in the game for actual gear this time, including the ability to upgrade existing weapons. It seems possible to use the same sword from the start of the game to the end, assuming you keep upgrading it often (if you wish).
You will be able to customize individual components of a weapon, such as hilt or blade.[5]
There will be an option to toggle helmet visibility. This setting is "global", so presumably applies to your companions. Possibly even NPCs.

Central Party Base
Skyhold Keep will serve as your "party camp" location, as well as the base of the Inquisition. It is at least partially customizable.
All of your companions will be there (like in DA:Origins), so you don't have to run around the map finding them (like in DA2).
There will also be several others keeps/castles/strongholds you can take throughout the game to serve as a strongholds for the Inquisition. It is unknown if these will also function as companion-interaction locations, or if you will have to return to Skyhold for that.

Important NPCs and Characters
Here's a list of characters, and what (if any) connection they may have to DA3.
"Old God Baby": It has been confirmed that if the ending slide of Origins did not mention her being pregnant, Morrigan does not have a child.
Flemeth: She will be present in some capacity, but no details are known.
Alistair: Steve Valentine (Alistair's voice actor) tweeted that he has recorded lines for Alistair in Inquisition.
Anora: Mika Simmons (Anora's voice actor)
confirmed via Twitter that she will be back as Queen Anora in Inquisition.
Bodahn and Sandal: Bodahn mentions at the end of DA2 that Sandal has been offered a job in the court of the Empress of Orlais. Given that fact, and the fact that they are sort of iconic DA protagonist hangers-on (like HK-47 and T3-M4 in KOTOR), they will likely (but not confirmed) be appearing in similar roles in DA3.
Duncan: He's dead. Get over it.
Lambert, Shale, Rhys, and Evangeline: These characters all played major roles in the novel Dragon Age: Asunder, by David Gaider (A good read, if you have not already. Highly recommended). The novel seems to be setting the stage for DA3, so it is likely that at least some of them will be in DA3. In what roles, if any, is unknown at this point.
Empres Celene, Duke Gaspard, Ser Michel, and Briala: These characters all played major roles in the novel Dragon Age: The Masked Empire, by Patrick Weekes (Another awesome book). The novel also seems to be setting the stage for DA3, so it is likely that at least some of them will be in DA3. In what roles, if any, is unknown at this point.
Maevaris: She was introduced to fans too late to be included as a companion, although there is more to her story. She will likely NOT appear in DA:Inquisition, but likely in future products.
The Warden, The Warden-Commander, and Hawke: The previous three (two and a half?) protagonists may yet play some role in DA:Inquisition, but nothing has been confirmed or denied. Considering their seeming relevance at the end of DA2, it seems likely that they will at least be mentioned. It has been confirmed that the Dragon Age Keep (the import system) does NOT track visual appearance, so that clearly throws some wrench into that.

Mods/User-Generated Content
There will be no toolset. It's possible there will be an "overrides" folder for simple mods (such as cosmetic ones). No word on this yet, but it looks like mods will not be well represented.
