Favorite Starter kit: Vanilladept. Really opened my eyes to how nasty biotics really can be since I'd never used an Adept in SP (2 vanguards, infiltrator)
Workhorse: Krogan Shaman. Up close & personal and still has great power synergy.
Favorite overall: Asari Valkyrie Sentinel. Tech Armor + Asari + proficiency with a wide range of weapons = 
Honorable mentions: All Drell, Smash Bros., Fury, Quarian Male Engineer
I'm nostalgic for the Human Soldier. I used him and the Salarian Engineer a lot when the game first came out.
This was my typical build for him. By no means optimal now, but remember a good deal of the weapons were not around yet and gear wasn't available either.
It was still fun to play though.

Starting Class (demo): Human Engineer
Starting Class (full release): Drell Adept. I annoyed more than a few people with the old reave sound, I'm sure. I moved over to the Justicar once she came out though. I just loved Reave!
Workhorse: It used to be the Justicar and Male Quarian Engineer. I'm pretty comfortable with just about any class at this point (I only hate the Talon Merc on 90% maps since the only aspect of him I find fun is laying traps), but the GI and vanilla Human Adept are my most trusted kits.
Favorite: N7 Paladin with a CSMG, cryo shield, and no snap freeze (for maximum tank). Far from optimal, but trying to make good use of that shield on platinum is just a fun challenge to me. Sucks that only the heavy melee reliably primes cryo explosions and that the armor de-buff is broke, but watching entire groups of cannibals freeze themselves by meleeing my shield just makes me laugh more than it should (freezing phantoms is a nice plus). Works completely fine on platinum, it just takes good judgement on positioning. I guess how quickly it punishes mistakes and the close range fighting with a shield reminds me a lot of dark souls, which I love.
The game seems so different from when I started that it is hard for me to pick a starting class. Also was it only vanilla humans that you start with?
Starter: Human Engineer, although Human Soldier would be a close second. Initial weapon selection is the difference here.
Workhorse: Turian Sentinel because he does everything.
Classes I like the most: I will combine favorite and runners up since I don't have a clear favorite. Human Soldier, Geth Infiltrator, Geth Trooper, Turian Sentinel, Drell Adept, Krogan Battlemaster, Drell Vanguard...
Also was it only vanilla humans that you start with?
Still is, although normally the starter gift packs contain one uncommon or rare character. (I got an AIU when I started a PC account last week.)
My favorite starter class was the human engi. Course, that was back in the day when combat drone was even better than now (still a great class though).
My workhorse class as I was getting my manifest going/learning to play, was the Salarian infiltrator. Top tier DPS and good survivability with energy drain.
These days my all around favorite is really every class who isn't the turian saboteur. He's the only one I've never had it in me to enjoy. Besides him, every class is fun in its own way.
Favourite starting class: Human Engineer. My last playthrough in ME2 had been with Engineer, so that was the class I first picked in ME3 demo. I loved how it worked in ME3 and used it a lot in early days - and I still do.
Workhorse class: Geth Engineer. I've used it probably more than any other class. It's probably the class that I'm most comfortable with facing any danger out there.
Favourite class overall: Asari Adept. I love the way asari play in general, and the adept is the most fun of them all. I love how every power serves multiple purposes and how you can combine them and weapons fire in so many ways.
Honourable mentions: Quarian Ice Queen, Huntress, Frog Engineer, Senturian, Assguard.

Favorite starting class: Human Soldier. You can't go wrong with HSol.
Workhorse class: Turian Sentinel, proved in practice.

Honorable mentions: (in order of classes in the menu) Asari Justicar, Krogan Shaman, Volus Adept, Krogan Soldier, Batarian Soldier, Human Engineer, Salarian Engineer, Batarian Sentinel, N7 Paladin, Salarian Infiltrator.
It will be revised eventually, when I get the chance to test all of my kits.
Favorite classes have continual changed until now from HS when starting to HE on silver to HE/QMI/HI/QFI/SI/QFE/GE/SE/Tsent/Demo/Shadow/Destroyer. Now i play engineers almost exclusively and with amp 1, incen 2, cyc 2 as a habit.
Favorite starting class: Vanilla adept (best biotic in game)
Workhorse: Jugg (a great jugg does a lot of work, keeps instant kill units of team, helps with obj, draws a lot of fire from enemies, ...)
Favorite overall: N7 Paladin
Honorable Mention: Turian Soldier, AIU, Ghost, GI, N7 Fury, Drell adept, Geth Trooper (I almost dont use my trooper anymore but he's still important to me becuz he's the char that I used when first doing platinums)

I love all classes and most kits. Unfortunately since I rarely (never) host I don't play vanguards nearly as much as I'd like to. Something about mashing Y 20 times and standing there like an idiot before dropping dead just doesn't do it for me. About the only character I really am not fond of is the Krogan Warlord. Running around with a giant hammer isn't for me, it gets boring fast. He's still a good character though.
