I've been trying to come up with a biotic assassin build but I can't decide on the powers it should have. I think its because whenever I think of the word "assassin", I feel the Tactical Cloak is compulsory.

I will be sticking to my 3-Power rule, where I can map three powers to buttons so I can instantly use them, thus restricting the amount of powers. Any additional powers are acceptable if they're an ammo power or an inactive power that you don't need to use too much (Eg: Fortification, Barrier, Defence Matrix).

What powers I'm looking for:
-a biotic crowd control
-a tactically used biotic power to strip defences
-possibly a grenade power
-If possible, try to implement Tactical Cloak or Tech Armour

If you are looking for "tactically used biotic power to strip defenses" you are not going to find one.

Flare could be used to strip shields I suppose, but unless you are using it on an infiltrator or Soldier, the cooldown may be too punishing. It can also be dodged.

Warp specced for power damage is probably still too weak to fully stip a Phantom Barrier or even the weakest armor on enemies. Same with Reave. And both of those powers are better suited for priming and detonating power combos anyway.

Biotic crowd control is easier since you have a choice between Slam and Lash bonus powers, and Pull, Throw, Singularity, and Shockwave are available on specific classes.

For grenades, you are limited to Inferno Grenades, and/or either Frag grenades (soldier), Lift grenades (Sentinel), Cluster grenades (adept), or Sticky Grenades (infiltrator).

Now, the other question is what do prefer by "assassin"?

If you prefer a 'backstabber' assassin type, then Infiltrator with Cloak is good for turning invisible and then walking in with a shotgun for close-quarter kills.

If you want a sniper assassin, then any class can use a sniper rifle, although Soldier and Infiltrator are the only classes that can kill elite shielded enemies with a sniper rifle without requiring Energy Drain/Overload to strip the shields first.

To me, your best bet is either:

1. Infiltrator with Cloak, Slam, Sticky Grenades, plus Disruptor Ammo (specced for damage) and the Kishock.

2. Soldier with Adrenaline Rush, Frag Grenades, plus Disruptor Ammmo (specced for damage) and the Kishock.

The Kishock deals a portion of its damage immediately, then the rest over time, which allows the Infiltrator and Soldier to one-shot kill most shielded enemies when using Disruptor Ammo.

Is this really an Assassin build if you are also prioritizing a Protection ability?

Having fewer powers is better, sure, but an Assassin works with several tools, not just one.

When you are asking for a "biotic assassin with crowd control and armor powers", you are really asking for everything in one basket. Assassin implies precision, while crowd control implies area of effect.

My first inclination was to make it an Infiltrator with the Bonus Power evolution, but if you're going to be all Assassin with the Sniper rifle, that's not possible.

My second inclination was to do a Sentinel with Tech Armor specced for Power usage, and Warp Ammo, allowing you to Overload defenses, prime a target with Warp Ammo, and finish it with Throw.

It's the closest you'll get to Thane's biotic assassin vibe.

I would approach this slightly differently. Something like this

Tactical Cloak - Damage, Recharge Speed, Bonus Power

Sticky Grenade - Damage, Max Grenades, Damage

Sabotage - Backfire, Recharge Speed, Tech Vulnerability

Stasis - Stasis Strength, Recharge Speed, Vulnerability

I would always take Liara with me to use her Warp Ammo bonus and spec her for Warp with Detonate, Expose and Recharge Speed

The ideal other squadmate would be EDI with Incinerate and Overload. Kaidan can be a good choice as well

Use Tactical Cloak, put an enemy into Stasis, close distance and blast with a shotgun, or a high damage pistol like Executor. Suppressor might fit the assassin vibe more but will require more shots (with upgrades, warp ammo and status bonuses - not much more. You can also add weapon damage bonuses from armor and intel)

Sabotage will be used on tough armored enemies like Brutes, giving more damage to EDI's Incinerate and tech explosions to her and Kaidan's Overload.

Hard to try and put all these priorities together without class modding. :)

Since it sounds like Cloak is a core power I am just going to go ahead and think aloud (so to speak) about an Infiltrator.

As pointed out above, there isn't really a biotic bonus power that is great at stripping defenses, at least not by itself.  The way I see it you have a couple options though:

1) Give up on the biotic part and use Sabotage or simply a Tech Squadmate to strip defenses.  Leveled up Overload can strip a Phantom's barrier in one cast, and can deal with shields on other units.  You can combine this with other options below, but it means you aren't shield stripping with a biotic power, if that matters.

2) Flare is the most powerful biotic ability in instant damage and would be nasty bypassing the cooldown with cloak and with the massive cloak damage bonus.  However, I would not personally take this approach as I think this is more a Michael Bay type character rather than what I would think of as an Assassin.

3) Go the DOT route with either Dark Channel or Reave under cloak and detonate with a party member.  I would probably take this route, just because Dark Channel cast from cloak seems pretty ninja to me.  Reave would be somewhat similar except it also gives you DR.

4) Go the "close range" biotic powers for a CQB style.  Slam or Barrier (detonate).  This might be the most fun option.

5) I don't like Stasis, but it makes some sense if you are "assassinating" targets, as it renders them helpless for easy head shots.  Doesn't work on armor, but that isn't a big deal.  Instantly drops Phantom DR, which isn't as huge as in MP but still helpful on a couple missions.  Bubble is useful at chokes, but again not quite as helpful in SP as MP.

"Thank you for all your loyalty and support!"

I've tried Red Caesar's 2nd option, because the only biotic bonus powers I have is Slam and Stasis.
Disrupter-Ammo is great for wiping out shields with a sniper rifle. I can either finish them off with more "Ms Archangel" shots or slam them THEN do "Ms Archangel" shots while they're stuck in mid-air because why not. Any armoured enemies, I can use tactical cloak to help me aim my sticky grenade at them without being seen and they grt most of their armour stripped off easily.
