I've got a couple moments, but I'll go with the joking one first:
Sandal. Maker's Breath, Sandal. What is the world IS that boy? All cute and adorable throughout the game, then you walk into Fort Drakon during the attack on Denerim and... holy hell. Darkspawn corpses everywhere and little innocent Sandal in the center o-o Granted, Sandal pulls the same trick in DA2, but even then it's full of wtf?
As for the moment that has me choking up every time I play the game (and there's video evidence of this, as I've Let's Played DAO) is the loss of Ostagar and Duncan's death. The music and cinematics were amazingly well done, and the tone of that piece, from beginning to end, from going up the Tower of Ishal to Cailan's death and Duncan's epic take down of that ogre. Great, powerful scene. I played a female Cousland rogue, which led to a a strong arch for me. I never even thought to spare Loghain; the second I got the chance, his head was off.
