In short, Lyrium is basically crack with magic steroids that gives Templars their powers.
That's actually unclear. If you remember Origin, Alistair says that lyrium isn't really connected with templar powers and it might just be a way for the Chantry to control them. That always gave me the chills and I'm glad they decided to address the issue in Inquisition. It would have been easy to forget it and just go with the "bad lyrium" issue, overlooking the effects of the "good one".
