In DA 2, it's mentioned that the Warden is one of the Hawke family's cousins, and Cullen will mention an Amell Warden to Hawke as well. Not sure if there's a reference in DAI

Yes, Daylen Amell and Hawke are related through his mom Leandra Amell.

Yes its the human mages birth and her/him being found to be a mage that prevents the Amells from becoming Viscount though they were considered the prime candidates and had popular support for their candidacy.

Its the main reason my canon Warden is a human mage. I liked the family connection very much. It wasn't intrusive, the bits of DA2 that mentioned the Warden as being an Amell weren't any more of a big deal than the ones for any other version of the Warden.

Yes, if you imported a mage Warden into DA2 and ask Leandra about her family she will mention how "The Hero of Ferelden was one of ours. One of my cousin Revka's children. The Circle took them all." So, not only is a mage Warden cousin to Hawke but apparently there were a few siblings and they were all mages too

And as far as I know the Trevelyan's are not related to the Amell's. Which was something I was rather disappointed in. I wanted to keep the greatness in the family

Yeah, didn't think so. Actually, misread the OP anyway. For some reason I thought he was asking about the Human Inquisitor even though he said Warden.

Technically they could still be distantly related, considering they are/were rather prominent Marcher noble houses.
