Does the rune type really matter, appart from against the monsters with the obvious resistances and weaknesses?

Runes of any kind will bump up the overall damage of your weapon. While some enemies will resist more of an element's damage than others, the increase to your total weapon damage will compensate for that.

That's just from my experience though.

But can they trigger any special effects, or are they just a damage boost were it doesn't really matter what rune you use as long as the monsters don't have resistances/vulnerabilities?

I think they may trigger effect depending of the element used:

ice : chill

Fire : Burn

Electric : Paralyze/shock

If that was the case, we should really have this clearly described on inventory screen - chance, duration, damage etc.

So far we are still guessing and making assumptions about game mechanics. I'd love this to be true as this would add extra depth to customizing weapons. At the moment all I see is just a straight damage increase.

Come to think of it, I've also seen random "asleep" statuses on enemies I'm attacking with staff spam. My staff has a spirit rune in it.

Yep!

Spirit = asleep (staff runes and riftmage skills)
Electric = shock
Paralyze comes from electromancer skills like Static Cage and Lightning Bolt.

Honesty i've never seen burn popup with fire runes.  I think the runes probably just add damage.  Either that or they dont have a status indicator for runes.  As for sleep thats a combo from shock + something else.  Don't think that relates to rune.

In SP weakening a shocked target puts them to sleep. Dont know if it is different in MP.

I do believe this applies to MP also.

In my recent build/promo for my AW, I would use Chain Lightening then Rock Fist (upgraded) to put enemies to sleep.

I also belive they trigger effects on crit.

Like the warrior ability Flow of Battle. Critting with your attack and your rune will trigger Flow twice, afaik.

That floating spider demon resists cold, the Terrors and Wraiths are weak to Spirit. Other than the pride demon resisting electricity, I don't think there are any other resistances/weaknesses worth noting.

Also, the Rage demon and Venatori Commander resist fire, Despair demon resists Ice.  I'm fairly sure, but not certain that the Venatori Spellbinders resist electricity, and the "floating spider" aka Fear, does NOT resist ice.

Yeah, no kidding.  "Robes don't offer as much protection as you might think."  Uhh, yeah, thanks, I never would have guessed that robes provide less armor than plate...   "The Avenger is deadly on full auto." Right...
