I recently broke it off with Josephine and began pursuit of Sera. I spared the noble in her personal quest, which resulted in a big argument between us in her quarters, and have been generally lenient with other judgments. Have I effectively lost any chance at romance with Sera or are there opportunities to make amends later?
After my disappointment with the Josephine romance, I'd rather not even bother if there's no hope.
I think you're still good, it will just take a while for the romance to kick off because you now have to spend time getting the approval back up (which isn't hard to do, don't worry).
Or at least, I could still have my character romance Iron Bull after sacrificing his Chargers. While of course it doesn't have to mean the Sera romance treats approval the same way, I'd say you can just go ahead without much trouble.
Finally i made to the quest where u have to talk to everybody for a present to sera - well - it doesn't work - i spent 3 1/2 hours and always it stucks at the last person to talk to - traveling and return brings the quest to a point that i talked with everybody, but no interaction with sera. i remember, that there was a interaction "go to my room" (or something else) but it was like years ago - no options available - only kissing, talking to her and so on. did i messed up with that option with the room going earlier? cause there was nothing happening - i thought it would be the night of my life, but nothing happend. so i just ignored. to go back to a savegame with that option, well - i'd rather skipp it, must be like 20-40 gaminghours ago - no thanks.
i talked to sera after completing adamant and had to cut through many dialogs, and even after - no options - no jumping into bed or else such kissing.
anybody has an advice?

Edit: Problem solved - i restarted the game and everything was alright.
