I'm not sure if this is the right forum for this, none seem to match specifically, but I was wondering, I've just started another playthrough as a Mage this time and planned to specialise as a Knight Enchanter. Is it possible I can bind my Spirit blade to the 'R' Key whilst retaining the left mouse button as the staff attack? Or does changing the key-binding also change the mouse control?

You can bind it however you like. I've had R bound to healing potion all along and left mouse functions as normal.

Yep. Most keyboard keys should be fine for re-binding.

If you have mouse thumb buttons, I'd recommend mapping one of them to spirit blade. You may have to set them up in your mouse control software to use keystroke assignments, though. Bioware didn't provide mappings for mouse buttons beyond the left/middle/right for some reason.
