Just wondering.  About to put one in my 2 handed weapon, just not sure which one to use.

Corrupted ones I think, iirc they affect all beasts (that would include dragons), humanoids (includes red templars) and anything else alive. Only undead and darkspawn are immune. I could be wrong though. I do like making a demon-rune equipped weapon for the Fade and Cleansing runes when I know I'm mostly fighting darkspawn. I'm sure someone will give us the straight dope.

If corrupted are as effective as it seems, I think there should be negative repercussions for making heavy use of red lyrium.

Lightning Rune, since it applies the most useful debuff in most cases. Frost Rune is decent if you are a ranged character relying on kiting.

Corrupting Rune doesn't affect dragons or red templars. It affects only regular humanoids (Venatori & bandits) and regular beasts (bears, wolves, giants, lizards, etc.). Enemy-specific runes are fairly straight forward - each covers only 2 enemy categories, except dragon rune.

Generally speaking, you will only ever run into difficult fights against Venatori (Corrupting), red templars (Cleansing), undead (Demon-slaying) or demons (Demon-slaying). Each is fairly situational and usually region-specific, but since Demon-slaying covers two such situations, and demon Rifts are in nearly every region, I'd say Demon-slaying is the most versatile among enemy-specific runes.

Finally, and this is pretty big since you are talking about a melee character. Enemy-specific runes give bonus weapon damage based on the weapon's original element. This means that a 2H sword would deal bonus physical damage, which is mitigated by enemy's armor and any other reduction. Elemental runes on the other hand bypass armor entirely. The difference between a 25 dmg elemental rune and a 50 dmg enemy-specific rune is smaller than you'd imagine when used by a physical damage dealer.

Spirit for Staff users of course, its the best "general" all around DPS increase for those with a staff.

I would go with lightning otherwise.. granted I can't seem to find a Superior Lightning Rune myself.. so.. meh
