This seems like it could cover a wide range of things, has anyone tested it? Is it only vs animals and humans (including the other races)? Not Dragons>?

Yes, it is only against humanoid creatures and beasts, NOT dragons. Dragons have a specific category appositely for them (dragon-slaying).

So effectively that would make spirit runes the most effective across the board?
That depends on how you're defining effective. It's a niche rune since it can only be used on a staff.

Edit: pretty sure red templars, darkspawn and dragons have their unique runes. Also elemental runes just do 1 additional damage if the enemy is resistant against certain element. Spirit rune is probably the one that works against most enemies but these can only be applied to staves if i am not mistaken.

Yes, Red Templars and Darkspawn = Corrupted. I think for Dragons it's Dragonslaying Rune.

I also noticed the rune damage only seems to be added to normal hits, not your special moves.

I am not certain but i dont think it works against red templars or darkspawn, which could be called humanoids. I think it just works against bandits and beasts.

Yep, "humanoid" was a bad term on my part. Didn't think that also Red Templars and Darkspawn can be considered such. For humanoid I intended humans, elves, dwarfs and the like.

So as I understand it:
Cleansing: Damage vs. Darkspawn+Red Templars
Demon-Slaying: Damage vs. Demons+Undead
Dragon-Slaying: Damage vs. Dragons
Corrupting: Damage vs. anything else not listed above
Frost/Fire/Spirit/Lightning: Damage vs. everything not resistant to respective element.
