Do points of interest ever get taken off the map?  Whenver I try to go to one of them I find nothing there?  Are they possible quests or something?

If you are talking about the triangular landmarks, you have to claim them (interact with the glowing banner). Then they become an X on the map. The X's stay on the map permanently.

She's talking about exclamation marks that don't turn into other map features within range. Those are quests that might need to be triggered in some other way.

If an exclamation doesn't turn into another map feature within range it's a quest that needs to be triggered somehow. The fact that there's a marker means its in your journal somewhere... follow the clues.
