If we are on the topic of favorite lines, and Awakening Anders....
Anders: Pull away entirely?! That's madness! I hate the Chantry oversight as much as the next mage, but they can't just decide to leave! That's a recipe for disaster.
It's only a good line from a certain perspective...

The line the Inquisitor delivers after Here lies the abyss:
"Warden Loghain is dead, thanks to all of you! He alone stood up against Clarel madness, and if it were not for him you would all be dead, or worse, working as servants to an agent of the blight. And how did you repaid him? By branding him an traitor!"
The rage and disgust in the Inquisitor voice is almost tangible.

Iron Bull: When you do, make sure you describe the masculinity right. Cause this isn't just endurance work -- there was a lot of strength training to get here. You wanna use words like "rippling" or "ripped". "Ripped" is good.
Varric: Hmm... The Iron Bull's belly is prone to rippling after every meal. He rarely wore shirts as they ripped under the strain.
Iron Bull: That hurts, Varric. That's hurtful.
When I heard that banter I think I laughed so much a bit of pee came out.

I didn't like DA2, I played sarcastic Hawke. :| Sarcastic Hawke is the only thing that kept me going in that game. Still a ****?

Don't know if this is pinpoint accurate, but when you ask Sera about where she grew up. It goes something like:
Sera: "Wherever? South, north, wherever I want!"
Inquisitor: "Oh, you're from wherever? I'm from north wherever. Oh we had fun on local street and/or in local tavern."
Was the first joke that made me laugh in Inquisition. The Warden and Hawke had better lines now and then, but I liked that one.

In DA:O when in Lothering Morrigan and Alistair talk as you go over the small bridge. They are talking about how Alistair liked to follow instead of lead, and by the end of it Alistair says one of my favorite lines.
Alistair: 'Couldn't you crawl into a bush somewhere and die? That would be great, thanks.'
Alistair says some great things, tbh.

The way he delivers the last part was probably the funniest thing I've heard in a while.

I have mentioned my favourite in all Dragon Age (from Awakening Anders), but my favourite in Inquisition is in the scene where Fiona says how she joined with Alexius. Vivienne just says, "Fiona dear, your dementia is showing."

Cole: "He's very angry that you took his mages." Cole's minimalist exposition is so much better at setting up a sensation of fear in In Your Heart Shall Burn than Dorian's far-too-long exposition, and he really nails that line.

Seriously. Aggressive Hawke and Sarcastic Hawke are like two different characters and they are both so very amazing.
