I like the dialogue with Blackwall when you first meet.
Quizzy: (something along these lines) "I'm finding hard to see any use for you. What exactly can a grey warden do to help?"
Blackwall: "Save the effing world if pressed."
Instantly gave me a flashback to DA"O and I smiled a little.

Right from the start, almost the first time my Inky spoke to Cullen I knew she was going to romance him. From the first time she flirted with him and that cute little clearing of the throat thing he does when he is a little embarrassed/shy and that smile!
Vivienne is probably one of the best characters in DAI, if only because of the number of people who react negatively to her.
Mary Kirby must be satisfied...
As Sera eloquently says:

Actually, if I'm not mistaken, Sera's exact words were "She's a b!tch. And she knows it. She better."
That one line immediately made me doubly appreciate Vivienne, and triply appreciate Sera at the same time. Sera's bluntness and her effective reflection on Vivienne's character make them so interesting.
I honestly wasn't sold on Sera at the time. She was funny, but her face was ugly on first close-up, because I prefer characters to have realistic faces that aren't too realistic. Plus BioWare didn't fix her god-awful fringe like I'd hoped. Fortunately, banter with Dorian explains why her fringe is so bad.
That said, Sera was always going to be one of the 3 characters I knew I'd romance.
Vivienne for wittiest lines, hands-down. Cassandra for her strength of character.

I was largely indifferent to Leliana in both previous games, but her crisis of faith and resulting personal developments made her one of my favorite characters in the series, right at the top, after Morrigan. Feels more human and less in the clouds.

That Cass line for me too. I was like... 'Wow.. that's him told'. 
Dorian: 'I am being clucked at by a hen, evidently!!' I had to pause the game, so funny.
I enjoy when Cassandra says to Roderick in the start when you first go into the War Room:
"Have a care chancellor"
The intonation is all 'CHICKITY CHECK YOSELF BEFORE YO WRECK YOSELF'.
I crack up at it.
Cassandra is my hero!
