The quest, Where The Druffalo Roam seems to be broken. When I go over to get the animal, it's not there. There was a couple times before that the animal would spawn but wouldn't follow you or if it did it'd get stuck IN a tree or rock and you wouldn't be able to complete the quest, even if you reloaded the save, so yeah I think there is an issue there.

patch 5? Havn't ever noticed it just plain not showing up at all on early patches.

The problem with it getting stuck IN things and not being able to complete the quest started happening before patch 5. As far as it not showing up at all that has only happened to me since patch 5.

Have you wandered around a bit and come back? when I've done this quest before (pre-patch) it wasn't at its spawn point, I left the canyon through the other side, came back in and it was there.
