Now that we are pretty certain that it takes place in the Andromeda with this ark, its not unreasonable to assume that the mission was launched after the start of the reaper invasion, and before the possible curing of the genophage, if the devs want a clean slate.
If we accept these premises, then we can be fairly certain about who cant show up in the the next mass effect, namely, everyone who is seen or heard from after the genophage, which would include the entire main cast along with many others. Now, it might get a bit blurry since there are a lot of "war assets" that are effectively helping but doing so off screen, so that might also rule out certain people, as well as npcs who stand around on the citadel but dont have any real new interactions past the genophage. Also, if bioware wants to truly have a clean slate, no npcs can be inlcuded whose lives have at some point before the genophage been in the hands of the player.
With that said, who from the OG could potentially be in on this andromeda expedition? I have trouble coming up with any significant ones. Unfortunately Conrad would be out, as would Kal'Reegar (actually, the entire ME3 quarian cast) and Shiala as well. Problem is that these prerequesites would be quite harsh, and it doesnt help that the citadel dlc can only be started after the genophage, where the most of the main npcs appear. Problem is if they relax the "pre genophage cure" conditional, then they are retconning, and then everything else is suddenly on the table. Not necessarely bad, but counterintuative to setting it in another galaxy in the first place.
