I personally hope they change up the cast roles a bit. Like, instead of a funny piolt, i would rather have a stone-cold hardass this time around, with a chip on his shoulder or like...no sense of humor...or super dry wit...something to make it feel like a different ship. Or maybe even two pilots who constantly disagree or have a slightly antagonist relationship...for laughs, but not because they are trying for it.
I would also love to have a batarian mechanic. I don't know why...maybe guilt for offing Cathka? And if she were female, that would be sweet.
If exploration is the theme as has been suggested, then we need a scientist. I would love it if it were a geologist (cause that's what I am 
), but I assume a biologist would be necessary considering food and ecology would be super important to nail down on each world. But I would like less Mordin and more Liet Kynes!! 
For humor, I would love to see a comical krogan. Maybe a bit of nerd who abhors violence, but can cook someone with his tech powers if he needs to.
Still...Bioware has to be careful they don't change up the formula too much. I still cringe when I think of DA2 and the "forced complexity" of the characters. Ie, a healer BUT...he is an abomination! A sweet little elf BUT...she is a blood mage! Ugggg...what trite nonsense that was!
As long as the characters are complex and, as others have stated, not there to worship me, then I will be happy.
Also...a tech expert that was a volus would be so cool!!! I want a volus on board soooooooooo bad!!!
