Maybe the PC just turns into a ghoul throughout the game. Mortality problem solved. Absolutely no downsides. Yup. No downsides.

Eh, who knows? Trailer don't aways show what actually happens in the story. Maybe the prologue have us playing the antescesors of the protagonist, and this is the game over cutscene if we fail to enter the vault before the bombs drop.

Quickly reviewed the trailer scene by scene; a few interesting things pop out:

Ant Egg clutch near crib?

Captain Cosmos Poster in distance at about 1:40.

Ghouls coming at the PC look significantly more aggressive...knocking clutter out of the way to get to you...more like Reavers.

Nuka-Cola vending machines completely revamped...or are expanded models.

New Board Game? -- "Blast Radius" (at the beginning).

New Pre-War clothing models

PC looks like he is emerging from the Vault vertically. Looks almost silo-ish. Substantiated through several scenes.

Mirelurk? At 1:34. If it is, it's a new model.

Deathclaws confimed. But are they counterparts to their tough as balls cousins in the Mojave or their wimp relations in the DC wasteland?

“Chem-I-Care” – Chem Vendor?

“Diamond City” – Diamond City Surplus, Chem-I-Care, Power Noodles, Swatters Custom Baseball Bats Store, in trading area. Most likely that's Fenway Park in the background. Also saw baseball player statues.

Looks like Paul Revere’s Statue at 1:56.

“GNN” poster at 2:02. Shows radio tower and lightning bolts…Galaxy News Network? Eric Todd Dellums is supposed to reprise his three-dog role as has been rumored, and he has teased on his twitter account. May also confirm where this game falls in the timeline.

New Nuka-Cola Posters and clocks.

Skill Mags? Well at least new magazine covers.

Minigun - Covered in background in the garage with the power armor. You can only see the barrels. Laser Rifle on pegboard in the background.

Looks like a suppressed hunting rifle of sorts leaning against the bench outside the power armor garage.

40 + new models/retextures visible of older FO3/NV objects that still look like themselves.

I'm curious what weapons will be in the game. My favorite has been the deathclaw gauntlet.

October release seems likely though, the dlc schedule is whacked. Seeing as this game has probably been in production over 3 years it is safe to say its almost finished. I don't think we'll have a forced male protag, the voice part i am not sure about though. All the more reason to just wait for E3, they have said they will share alot of details there.

October is possible yes. The DLC thing and PS3/360 releases are impossible.

Not really sure where I would stay on that. All the fallout's PC were not voiced and it worked because they were blank. You could up your own background. While a predefined character like Shepard would work, I don't know about a fallout blank one :/

I'm not really sure either, but it could be a great addition. I certainly enjoyed Dragon Age more with a voiced PC. Probably a minority opinion, but there it is.
