I bought the Extended Edition of BG2 during the Steam Summer Sale. I know there's the big part in the middle where you go to Spellhold and have to get out through the Underdark, and how you basically don't get back to the World Map for a while. My question is: Do any of the quests available before this point become unavailable after you get back?

Almost all quests in Chapter 2 can be delayed to Chapter 6, if you want to (Chap. 6 is when you get out of the Underdark). The rather obvious exception to that is the main story line part, e.g. to side with Bodhi or side with the shadow thieves. Also, lots of personal quests have timers (do in 10 days or less, etc.) - you want to be sure those aren't open before you leave for Spellhold.

My recommendation is to play as many of the quests as you can in Chapter 2. The reason for this will become apparent if you roleplay your story. While there is an argument for rushing along the main story (since it's supposed to be urgent) you can use the excuse of needing to build your power to take on your enemies to delay. When you hit Chapter 6, the urgency is more pronounced than ever and delaying to do random quests seems less likely a scenario when you've already grown in experience.

There are, I believe, eight major sidequests in Chapter 2, and from a roleplaying perspective you only need to complete one or two of them before you meet the requirements to start Chapter 3. (I think you can still complete those quests during Ch. 3 since you'll still be in Athkatla, but the Spellhold section you're referencing occurs in Ch. 4.) I've contemplated leaving a few of them until Ch. 6 before (for reasons that are too spoilery to get into), but the companions, treasure/equipment, and XP you gain from them is simply too good to pass up. Also, as Kevin Lynch says, it may not make much sense to do all of them in Ch. 2, but it makes absolutely ZERO sense to waste time completing them during Ch. 6. You're welcome to put them off, of course, but I would advise against it for both story and gameplay reasons.

Regardless of what you choose to do, welcome to the wonderful world of Baldur's Gate! Remember to always go for the eyes!
