I find it interesting that when you venture into the Deep Roads, Deep Stalkers seem unaffected by the taint yet spiders are.

I think dragons will factor into discovering the nature of the Taint and how to cure the calling. We've learned that they are highly resistant and can even quarantine infected parts of themselves somehow. And Yavana calls them the blood of the world or something.
And throw in the titans, too. They might have a role. Can't really think of what now, but maybe the Taint is a blood disease one of them had, and then it spread.

I still don't know where this came from honestly,those dragons of the Architect in the mine were corrupted just like the one of Corypheus.
The taint is not a problem anymore since Avernus maage to use it to improve the lifespawn,the problem is the calling.

Dragons are resistant but not immune to the Taint. So it's more difficult for them to be completely infected, even if they get some of the Taint in their system. But yeah, that wasn't mentioned in DAO so it's a development that we learned in DAI. Whether that was always planned or a retcon is unknown...
I think it's easy to determine, even if the taint isn't fully understood, but I understand the taint to be physical and magical, but the magic emanates from the physical. Here are the possibilities:
1. It's a medicinal cure. It is a substance that purifies the taint physically, which disconnects the Warden from the taint magic.
2. It's a magical cure. It is a spell or magical process that cures the body.
There are then possibilities that arise from these options.
1a. Plants, such as the flower that grows in the wilds that can cure a mabari.
1b. Chemicals, such as those mixed under herbalism or even poisons. Maybe there is a certain poison that kills the taint. If the body can survive this poison, it could effectively be a cure.
1c. Extraction. I can't really see this being plausible without magic, but it's possible that there is a way to extract the taint from the body completely, like drawing venom from a wound.
2a. Cleansing. Runes with the cleansing attribute have an effect on tainted creatures. Maybe they can be modified or amplified to destroy the taint within a Warden.
2b. A spell of cleansing. A specific spell that cures the body of the taint.
2c. Modification. One way to cure the taint is to restructure the body to be absent of the taint. One way to do this might be to go into the Fade physically, but somehow reform the physical body they using magic. If time magic is involved, one might be able to restore the body to an earlier point in time, to before the taint was contracted.
There may be a few other options, but these are the most likely, I believe.
Now there is the matter of what kind of cure would exist out to the west where the blight has not reached, and how accessable is this cure?
I think the only kind of cure that can possibly exist in the west is a panacea, or cure-all. Meaning, it doesn't cure the taint specifically. It cures just about anything and everything. This cure is likely physical and magical.
As for accessability, I wonder if this is some kind of potion that can be carried, or if it is location specifc. For example, this cure might arise from some kind of natural source in the earth. Or it might be a specific temple with magical properties. In which case there is no way to transport it. This would mean Dragon Age could continue with the storyline of the Calling and the taint affecting people and possibly killing them, but also have an "out" for special characters such as the Hero. And then instead of being sent to die in the Deep Roads, the Wardens could have the choice to go west and experience the cure.
The other possibility is to have this as a potion, which can be replicated. But in this case it would reduce the taint to something of a debuff that requires an inventory item to reverse each time. Very RPG, if you ask me. Plus, if this is a cure-all, then that means it could cure many things, from poison status, to say... infertility perhaps? *Cough*Anora*Cough*
