So, is Shepard's survival in high EMS destroy now canon? Or was this ship launched with a nice little speech from Shepard prior to the Crucible choice?
It's probably not meant to be taken literally. It could easily just be Shepard sending a last farewell to us, the players 
I bet that's all we're getting today. 
If that message is canon then the Ark most likely left before the end of ME3. That plus some of the things she says like "we have no choice but to try...for our fear of what should happen if we don't" make it seem like they're indeed running away from the Reapers. And like someone already noted "Ryder" might be the new PC or a main character at least. That dogtag was put there deliberately. That's all I got from the video.
Edit: I guess that video also disproved the theory about using a Collector cruiser/Black Ark to get to Andromeda.

More likely that the tempest was launched before the Mass Effect 3 ending.
I do hope this is the alliance reaper backup plan, because daaaaamn did it annoy me, that noone anywhere really did anything about the reaper threat. Made all the races look incompetent. I mean not even some kind of concept planning of a sovereign class ship killer anywhere... That they didn't have the time to get it ready? Fair enough... but no preparations of any kind were really shown, other than vague mentionings...

It always made way more sense to me that the mission to Andromeda left some time before the end of ME3. Seems that's right.

If the Ark leave post-ME3 ending then it has to deal with just as much variables (dead/alive Krogan, Geth, green synthesized people or not-green peope) as a post-ME3 ending game set in Milky Way. Zero point switching galaxies if the Ark leave after, it was obvious from the start. Some people still in denial of course hoping for Destroy becoming canon lol

Well , I think the time Shepard make the speech take place between ME1 & ME2. because Shepard is a Hero And after ME2 they lock Shepard on the ground, I don't think they like to use Shepard's speech to encourage people on that ship.
Basically the voice over is Commander Shepard sending a speech to the crew of the Ark who are about to embark on their journey to Andromeda. To me this pretty much confirms that the Ark leaves the Milky Way during the events of the Original Trilogy, my guess is probably at least sometime between ME2 and ME3. It also confirms that the majority of the people on the Ark are Human. Where the other council races tie into this I'm not sure.
I am wondering if it is even between Mass Effect 1 and 2, for I wonder with Shepard's situation at the end of Mass Effect 2 and beginning of Mass Effect 3 if they would allow them to address the crew.

I loved this trailer and I have it running on repeat right now in a playlist I but how chilling would it be in a way if we get set up in Andromeda or at the end of the game and if we some how make contacts with the folks back home just before the reapers hit

What would've been nice is if the ship left during ME3, they showed a few reapers in the video as the ship flew by
