It would also make sense from a lore perspective. A subset of humans was launched out of the Milky Way in case the Reapers win the war, so that at least the survival of the race is guaranteed.
I think it'll be during ME3 to be honest. It doesn't seem to me it could happen during ME2, and after that it was short time before Shepard got arrested.

And he was wrong to me. Like the whole time in ME2, you search for colonists. "Fight for the lost". Desperately trying to give humans a fighting chance in the world. You weren't there to embody ideas, but to preserve tangible, material welfare (albeit, it was a failure). Only in TIM's mind, was it all about ideals. It's why he found it so easy to abuse humans themselves.

So if the Ark did leave during the events of ME3, they wouldn't have reached Andromeda before the end. Which means, the people on board the Ark would probably have been able to see the shockwave from the Crucible/Citadel as it covered the entire Galaxy. There must be some speculation above the people onboard about what happened.

Shepard knew the Reapers were coming and that they were essentially an impossible foe. That message could have been recorded any time after ME1.
