No for the first three. Go with what you like and disregard the dumb comment up there. I cannot advice you on which side-quests to skip since I do them all. ;-) Tried to skip the shards once...ONCE. It didn't work out. Well, I do skip a certain desecration at times but will never, ever skip the Mire =D. I prefer doing the DLCs after the main story.
Finally going to play this game

1. Nope.
2. Elves get some minor extra stuff late in the game, but not so much that I'd recommend playing as one if you prefer a qunari. (Unless you do decide that you want to romance Cullen or Solas - Cullen is only into female humans and elves, and Solas only female elves.)
3. Go for Sera! Her romance is delightful - and playing a qunari is extra fun if you do.
4. The 'main' sidequest in each area will be introduced when you first go there. You should probably finish those. You can skip most of the other side quests if you want, and especially the requisitions. Those literally never end. Just tell the requisition officer you're too busy to collect crap for her.
5. Do all the companion quests you can! Some require raising approval to a certain level before you can unlock them. Also, if you're only planning to play once, I strongly suggest recruiting everybody.
6. The Descent scales with your level, but Jaws of Hakkon is high-level stuff, best done right before the final quests (you'll know them when you see them) or after you finish the main game. Trespasser can only be played after the main storyline.
7. There is a bit more qunari armour now, especially since you have the GOTY edition. Have fun dressing like the Arishok.
Pretty much covered everything right here tbh without going into too much detail.

^oh yes, I personally don't look on requisitions as quests so those I def skip a lot.

Surely Cullen's romance is just as important to the lore as Cassandra's?
Can you take Cullen with you on quests?
And don't call me Shirley.

Unless you are one that MUST clean the Journal of all quests, accept a Requisition from each area, and leave it unfilled at some point. This allows the Player to choose which to do for RP reasons, and then gleefully ignore more requests at every camp site.
Also, when you clear a requisition at your home base rather than at a camp, you won't get a new one until you return to the area and meet the requisition officer again.

What, miss the goat?!The Fallow Mire is fairly disposable, for instance; not that it's bad, but it doesn't really advance the plot.

The Fallow Mire does nothing to advance the plot, but it IS a fun side quest and provides the first real look we've had at the Avvar.
Plus, y'know, Movran the Under.

Mosaic pieces 2 warden artifacts for blackwall ,loads of tier 2 chests, warden battlemage armor and an avvar agent is worth it, smallest map outta the starting ones too
