Some Questions I have;
Why are we going to Andromeda?
Who are we bringing?
What will we do when we get there?
Some assumptions;
Reapers
Mainly human crew, an AI, a few aliens
Find a planet to colonize then begin making good relations with the inhabitants of Andromeda
I'm really hoping that they just don't have another "Citadel" with a council where we try to prove our value, find and expose a traitor, then become the hero race only to find out about an even greater threat.
I hope that we get something new. What do you think will be the plot of ME:A?

I'd love it if we were just adventurers/explorers tooling around this new galaxy making friends and enemies while we solve other people's problems. The typical "looming threat" that drives most BioWare plotlines has gotten stale. One of DA2's redeeming features was the sort of unstructured conflicts that arose of just living and doing things. Each act in Hawke's story was driven by a different conflict, but it still all tied together.

MW colonists go to Andromeda on an ark to escape the Reapers. All contact is lost, they are on their own, OR they have a QEC that lets them actually talk to the MW and find out about the post-ME3 galaxy. Which would have no bearing on Andromeda's story, as they probably left before or during the Reaper War.
The colonists are exploring, looking for worlds to colonize for each of the major species, while also discovering Remnant tech and unraveling who they were. They think the Remnant are benevolent for some reason. My guess is the ark ship was constructed by the Remnant, and the MW colonists interpret this as Andromeda giving a helping hand to escape the Reapers.
At some point, they realize they were horribly wrong and (predictable plot twist), the Remnant have their own plans and it was actually a trap. They may even discover that the Remnant have brought other species to Andromeda in the past, like the Protheans or Inusannon, which would be a convenient way to reintroduce such species into the lore and narrative.
There will probably be a more player-centric quest in there too, like a quest to find the N7 brother of the player character who went missing prior to the start of the game while on an exploration mission. There will also probably be numerous conflicts with the Khet and other native species.
Predictable story. It's Bioware's way. I hope I'm wrong, because it would be kind of lame.

Why go to Andromeda? masseffect.wikia notes the following and this got me thinking:
"Very little is known about the origins of the Prothean race; even the name and location of their homeworld has been lost to history. It is unknown precisely when Prothean civilization arose and how long it persisted, but one of their communication devices discovered on the planet Fehl Prime has been dated to as early as 68,000 BCE. After achieving spaceflight, the Protheans discovered the ruins of a previous spacefaring race, the inusannon, and from those ruins learned about mass effect physics and developed FTL technology." "The inusannon were a spacefaring race that existed at least 127,000 years ago. Little is known about them...."
Would this draw your attention to Andromeda? Are the Protheans or the inusannon already there?
Just wondering
At the very least we know that there are no Halo Precursors there, and the Janus Key wouldn't help us either.

Nerds sure do love to complain about companies playing it too safe and putting out the same things over and over, but as soon as things change the sky is falling.

We go to Andromeda because one, BioWare don't want to have to work around their mistake with the ME3 ending and two, it's simply a new exploratory mission. Much like we do now on Earth, planning missions into space, trying to get the first man on to Mars, we as a species are never satisfied. We always need to do more. In Mass Effect, that would be journeying to another galaxy.
How are we getting there?
The Ark is a giant ship the size of a small planet, self sustaining and capable of making it through dark space. This ship will carry several billion people, made up from all the different races. This is so that the different races that make it to Andromeda are capable of sustaining a population.
When we get there, we send out survey missions to different planets to learn about the new galaxy. We are one of those survey teams. As we are exploring a new planet, we happen upon a new race indigenous to the Andromeda galaxy, and they capture us and take us too their leaders. With the unlikely help of someone from a new race, we are allowed to leave, so as to bring a message to the Ark telling them to leave or war will come down on them.
And that's all I can think off.
That's my story. BioWare, you better not steal it!

No clue but my hopes
- We are going there as a last ditch contingency from the reapers.
-Ideally it will be council based with many but not all the species from Milky Way
-Assume we’ll be colonising
Assumptions i'm making
No reapers there
Maybe an AI managing the ark life support whilst the majority of crew/colonists are in cryo, remaining managing the ark habitat once we arrive in Andromeda .
Pathfinder Forces of the 4 council races including human N7, technically under the authority of a council like colonisation committee, that is supposed to manage the colonisation effort but in reality there’s a lot of frontier do what they want mentality amongst the actual colonies.
Plot involving conflict with the indiginous remnant.

I expect we'll be searching suitable planets for all brought species, oppressing less advanced locals, getting beaten by more advanced locals and pursuing Absolute Power in Remnant ruins. Nothing extraordinary, but could be fun if they don't overdo with PG-12 morale lessons.

Well, to find new places for all the species of the milky way !
I want to bring all species with me ! a ship with mainly humans inside, no, thank you ! I like diversity, I like my alien crew and I'm already so glad a Krogan is confirmed ! I think the ship is a big multirace flying citadel.
And what will we do : I'm not sure where the story can take us... I just hope they won't copy/paste the story from the ME trilogy on anotehr galaxy. Oh, and no reapers ! I dont want to hear from them again. I'm also not really happy to know that Cerberus will be part of the game ...

Why? We're explorers. Want to see different areas.
Who? Humans and some aliens
What? Build colonies. Humans will be in charge. As it should be. Fight bad guys. Discover some ancient artifacts. Defeat bad guys. The end.
