There is a new vehicle mode where you have a driver, gunner, machine gunner, and maintenance/turret spawner/mine deployer. It's pretty fun and makes people like the Mako for once. Lots of customization and equipment to switch out on it too. A few months after release they are coming out with battle dome where people take their Mako and their squad into a PvPvPvPvP arena.
There are 2 new races (that I know of at least others may know of more). Yahg and Sirini (think Asari but all biotic class. Crazy powerful, but no fitness or ability to use guns and they also levitate)
Every character now has 10 abilities to choose from and spec into as well as various passives.
To add more depth to combat there are bonuses given to each class. An example of these would be "suppressed" for the soldier class where enemy effective range and movement speed is reduced as they are being fired upon and pinned in or around cover. "Perfectionist" a minigame exclusive to infiltrators that is available when in cloak where it is possible to 1 hit kill any enemy with any scoped weapon in you can properly hit the "Fatal T" in humanoid targets or the specific weak point in others.

A similar AI "dog" like the one from ME3 that stayed in the hangar now doubles as a companion and party member as you explore.
Lance Henricksen voices a damaged and dismembered synthetic if you are lucky enough to find it.
There is an RTS minigame for when you are growing and expanding your new base/colony. Heavy influence from Starcraft.
As you gain and bond with party members you have the ability to send them out and do menial tasks that you don't want to do, but still need to be done. As you do this the game offers the option to refer to them as "Inquisitor" during these times. You can also send them on a suicide mission where there is a 100% chance they will die. Some party members will object because of the "odds" but if you respond with "Where is Kaidan or Ashley when humanity needs them." They will go.
