It is probably a safe bet that the game will begin with the Ark departing the Milky Way, and probably an opening crawl or voice over that gives the background of the Reaper War for new players, who might not be familiar with the story of the first three games.

Yeah, I just don't see this game not giving players at least some sort of recap.

This seems about right. Only question in my mind is will it all be a cutscene or maybe some type of small playable opening, like Fallout 4 had.

I'm not sure. If this happens then there'll be a timeskip until we reach Andromeda/when our PC is starting his/her mission.

It has the main character asleep on the Express ship to Andromeda dreaming of the day he/she left the Milky Way. Then awakes to a loud noise. The ship is under fire. Everyone runs to their station, but at that point its too late. The ship is heading towards an unknown planet. Everyone braces for impact. Once the dust clears, the main character walks around making sure folks are ok and others with injuries are being taken care of. Opens the hatch to see outside. The title Mass Effect Andromeda shows up on the screen.

There might be some kind of tutorial horde mode mission for evacuation, to show that Ark is heading that far for a reason.

I'm only half-joking. Fleeing a vastly more powerful enemy, a lone human ship enters an unexplored region of space as the protagonist is woken from cryo-sleep....
(To be clear, this isn't a knock against Andromeda, or anything--it just fits surprisingly well.)

Of course, its way too early to say since we don't even know anything about the background yet but I could imagine something like this:
It starts with a view of the crucible construction project as we hear Hackett speaking with another officer (possibly the captain of the arc and an important NPC later in the game) in the off:
Hackett: "Are you sure this will work?"
Other Guy: "Nothing is sure at this point."
H: "What about the backup plan?"
OG: "Everything is in place, the councilors have been notified in person."
H: "Good. Absolute secrecy is key."
OG: "What about Shepard? You didn't say a word to the commander."
H: "We can't risk it. Shepard's too exposed. Let's just hope we will prevail."
OG: "Well, sir, as I said, nothing is certain at this point."
While this happens, we see the camera pan over to reveal that next to the crucible, the ark ship is constructed.
Then the little opening text that stays on screen way too short to actually read it comfortably appears:
"In the year 2186, ancient machines, known as the Reapers invaded the galaxy from dark space. As they swept through star system after star system to obliterate every living being in their path, the civilizations of the Milky Way launched a desperate attempt to safeguard their way of life. A giant ship was built, using prototype technology to traverse the vast empty void between galaxies.
Carrying delegations of every sentient species, it was called The ARK.
It's destination..."
Title screen: ANDROMEDA
(The Mass Effect logo fades in above with a 1-2 second delay).
The actual intro would start with the crucible leaving for earth and the finished Ark ship staying behind to hear word of the outcome of that battle. After the crucible is gone, the reapers attack and force the Ark to leave in a hurry. ideally, at this point, we take control of the protagonist and do some tutorial/prologue stuff, maybe fend off a husk/cannibal boarding party during the escape.

Haha, don't fret, it's not like he's coming along. But you can't argue that Lance Henriksen's VO is not the worst way to manage the hand-over from the old trilogy to Andromeda. After all, Hackett is pretty much the only one, high enough up the food chain so he could be involved in the project and who cannot be dead or is indoctrinated.

I hope it's like ME1 - explaining the premise (something about the Ark-Project/Ark-Contingency) etc. to let us seamlessly slide back into the ME-Universe (while forgetting about ME3-Ending-Disaster!)

ps: I booted up ME2 three days ago (had some time to burn and didn't want to buy a new game, so I burned through it once more (just finished Arrival-DLC as the last Quest (ok: I didn't do Firewalker this time around...but I played overlord for the first time (had Bioware-Points that were collecting dust already, so I spent some!)...was fun...but now...ME3? I don't know, I don't like that game (appart from the really fluid gameplay - compared to the clunky ME2 gameplay that gets you killed more often bacause you can't role to cover etc.)...that's depressing IMHO
(it shows how great ME1 and 2 are that I am still miffed about how they butchered/mutilated/raped ME3 (IMHO - I know a lot of people have forgiven/forgotten, but I haven't: Still holding the line (Extended-Crap-DLC did nothing for me, it doesn' change that Cerberus is totally OOC - even indoctrination doesn't explain this...the operatives may be extremists (like Kai Leng) but they aren't stupid enough to let somebody reaperize them! TIM would have trouble with operatives leaving in droves IMHO! - and it doesn't do away with the crappy ending!), even more now becaus Dragon Age: Inquisition didn't flash me (plays like a single-player-MMO and the removal of healing still pisses me off)...hell, they did worse here than ME3 in a way: THE ACTUAL ENDING IS A DLC this time around!) with the linearity (RPGs shouldn't be linear IMHO and they shouldn't discount your decision), the elimination of the dark-energy-plot-line and that FUBAR of an ending ("Artistic vision" my ass, stop smoking pot all day and start making good games again damned - unless EA is stopping you: If so - leave and start a new company damned (or try working for/with another big publisher like Activision-Blizzard (they at least have a good philosphy "When it's done" so no extreme stupid deadlines etc.))

I'm not saying they should do it--just that the completely unrelated intro to a completely unrelated game, that I went looking for to post as a joke, fits the bones of what we know about Andromeda's plot surprisingly well.
Personally, I want the game to start off with an older Asari - or Krogan, I guess; that'd be a pretty cool subversion of their usual image - teaching a class of schoolchildren about the Citadel races, the civilisation of the Milky Way, and the start of the Reaper War that destroyed it all--at least, so far as they know. Pull back, the class is being taught in one of these buildings:
Pull back further, and those buildings are revealed to be aboard this:

As it starts making its final approach towards Andromeda. Cut back to the classroom, and we realise that one of the kids in that class is named Ryder. (There should be at least one future squadmate in there, too.) Then time-skip. Think the Fallout 3 intro, but more, well, Mass Effect.
(I really like the idea of them going with a generational colony ship. I love **** like that.)
