They did talk of a Ryder family, what if the villain turned out to be your cousin? I could see some storytelling potential in that.

I took the Ryder Family to be his way of saying there different Ryder background stories. But I rather the BIG BADDY/BADDIES be a race that predates anything and all knowledge we know all about.

I'd be ok if you are male you get sister and vice versa. Sibling instead of cousin would be I wouldn't say more personal but pretty personal. If we can customize our sibling and leave parents out of it that wluld be tight. Just as Long as PC Ryder is N7 or we both are.

Or you can: push them off ledges, out of windows, electrocute them, stab them, burn them to death (with oil and/or lava), drown them, decapitate them or run them over.

I also have a sister so I would prefer a brother in-game but I bet we get stuck with the opposite gender.

Doesn't matter in Japan, in fact, many of Fire Emblem games have first cousin relationship as canon like Lucina and Owain.
