I'd prefer to have a prologue than say flashbacks. If there is tension between the crew or between Ryder and their family I would like to see it play out in real time rather than being told they had a fight sometime in the past. In lieu of burying the player under a ton of exposition they will probably have a short intro followed by a codex info dump.

This is just a good idea that needs to be followed in general. Show, don't tell. All movies do it. At least the good ones.

Would be nice but i think we'll have a prologue in which we are already in space, must likely in a shuttle heading towards the Ark. So yes, we'll be in the milky way but it's not like we'll be on a planet, imo, which makes it pointless in terms of interactivity since we are still on our galaxy because someone says so xd Walking on Earth or one of the other planets seems unlikely. Probably, all the planets we'll see are planets in the Andromeda galaxy

As someone else pointed out it could be that the reason for the Arks might not have much to do with the Reapers. The trailers have put a strongly stated about humanity's desire to explore and push the boundaries, it could be the Arks and the people going to Andromeda aren't going there ONLY just to escape the Reapers. Some people might want to get away from the geth, dark energy, the various governments and corporations that run the Milky Way galaxy, some want a chance to make money and/or gain political power that they couldn't get in the Milky Way Galaxy, some might just want to be the first of their races to see another galaxy, etc.

I'm betting that what we're seeing in the E3 trailer is an emergency departure from Earth orbit as the Reapers approach. We'll probably get a Hackett cameo and perhaps Anderson as they order the Arks to bug out immediately. Then Reapers show up and start blowing up everything in sight. This handles the cameos and explaining why we're in Andromeda.

That's actually not a bad shout. Acknowedge the previous trilogy and immediately set up the new one. Remove any arguments about how the last trilogy 'ended' because the crew of the arks will not know. Also places a high degree of urgency on the 'mission'.

The main menu will show the ark's interior and stasis pods, and when you click New Game, the wake up sequence will initiate. "We made it."

Traditionally, that hasn't been the way that BioWare has handled things, or at least in the lead up to them. They've typically shown things that they've already finished, which in every case I can think of, that's the prologue. That's why I say that "We made it" is at the end of the prologue.
Also another thing that kind'a concerns me is how much she's breathing. It sounds like she's out of breath. At first we thought that was just a side effect of the cryo, but what if her ark was actually attacked? That would put a double meaning on the words, "We made it." I'm guessing that's the case, as BioWare doesn't like to anything halfway.

Well first of all, in the last two games the "MASS EFFECT" title came after a period of intense action (the Normandy being destroyed and escaping Earth) but before the plot really kicked off.
So I think it'd be plausible to assume that the order of events would be Opening -> Mass Effect title -> Waking up in Andromeda and meeting the new characters.
We need something epic before we wake up, or we don't really get the impact/falling action of getting to know the Ark and its people. Heck, it'd seem a little odd to have the "we made it" line and waking up process otherwise; why not just cut to Ryder getting ready to undertake the first exploration?
Personally I'd love a Hold the Line moment against Reaper forces as the Ark takes off. Show their full devastation in an engine designed for environmental destruction, and show why members of the Ark are so determined never to return to the Milky Way.
