I doubt it. Varric seemingly has a huge thing for some girl from his past (not going to spoil it for you). Varric strikes me as the kind of guy who wants a family, not just a fling, whereas Isabela seems to be the kind of person who doesn't want anything serious.
Isabela and Merrill is, IMO a more likely pairing. I think Varric and Isabela are just friends.

Varric is already fixated on some married woman who is usually a country away at all times. It's a sad, unhealthy situation.

Isabela and Merrill?! Isabela treats her like a naive little sister, or a friend's naive little sister. Plus, there's never been any indication of romantic interest between them.
Isabela is pretty straight-forward. When she's interested, she lets you know. Ex: Fenris. With Varric, I think she just plays around; more so because Varric himself likes humor.

Check your messages in Gamlen's house after completing A Business Discussion (talking with Varric in the Hanged Man). One of them should start the Bait and Switch quest. Go to Lowtown at night and talk to the dwarf Anso in the market area and proceed from there.
