Happened to me, I ended up having to grab a different DVD drive from another computer and hooking it up to mine. That one could read it, so I got it installed that way.
You could try mounting an image with DaemonTools. There are instructions around here somehwere (I'll see if I can find them).
Finally, I've seen a report that DVD43 can interfere with reading the disc. If you have that program installed and running, disable it.
*edit* Found the instructions:

*edit* Found the instructions:
Step 1. uninstal all copies of me2
Step 2. put cd2 in your cd tray
Step 3. Useing deamon tools or a program like that make an image of the cd
Step 4. Mount said image of disk 2
Step 5. put cd 1 in cd tray
Step 6. instal, it shouldnt ask for disk 2 becuase you have it mounted in a virtual drive
Step 7. ???
Step 8. PROFIT!
Also
to make sure you've got everything you need your mass effect folder at
the end of the instal should total roughly 11.4GB if it's less you are
missing something smack yourself and start from step 1.

If you don't know what DVD43 is, then you don't have it. Daemon Tools is free, yes.
Don't worry about step one. If you don't have anything installed, just go to step 2. I haven't tried the method using Daemon Tools, so don't know how it works, but I've read it has worked for some people.
Failing that, try hooking up a different DVD drive to your machine. If that doesn't work or you don't have access to another drive, you might try updating the firmware for your DVD drive.
Failing that, about the only other option you have is to take the game back to your retailer and exchange it for another copy. Perhaps a new copy will work.

Oh, one other option: you can install the EA Download Manager and use your CD keys to register the game. This will allow you to DL the digital version from EA.

Heh, I don't know. Hopefully you've booked passage on the QE2 and not the Titanic...
