I think it was his passionate inflection that had me laughing for a good five minutes.

Samara's line paraphrased "Help me find my long lost daughter and kill her."
Tali's line "I did find something better. I've got/found you."
The moment when the Normandy flies out of the collector base, the music is going and the camera zooms in on the ship as the place goes up. It was so great. What a final hour or so...

For Paragon Shep, it's Tali's trial or Zaeed's mission.
For Renegade Shep, every waking moment if either this mixed liberally with puppy-kicking evil.

So I'm going for the heavy weapon achievement and I load up the tac-nuke gun for a mission. After I find what seems like a good spot to use it . . . I nearly blast my team to cinders. As we are all getting up off the floor Jack yells, "You ******!"

You don't headbutt the shaman, you headbutt Uvenk. The Krogan with the green crest. If you're renegade enough you get to battle him and kill him after Grunt's rite of passage. The Shaman is amused by all of these antics though, and takes a liking to Shepard.
Actually you fight Uvenk whether you're renegade or not.

1)"Sir Issac Newton is THE DEADLIEST SON OF A ***** IN SPACE!!
2)KROGAN POET!!! "Oh blue rose! Let your roots dig deep, into the hot and scorched earth of Tuchanka! For if our love is to survive, it must grow thorns sharp enough to pierce any hand which would uproot it" EPIC POETRY
3)One-shotting the Human Reaper with the Cain. "I'm sorry Baby Reaper did you just ask for a NUKE GUN TO THA CHEST CAVITY? I think you did!" Actual quote, lol'd for 10 minutes straight

Also killing a charging Krogan by punching him in the face with my shields gone, almost no health and no ammo left

Finding Garrus. It's a Crowning Moment of Awesome for the both of them in defeating all three merc gangs, and a Crowning Moment of Heartwarming for the actual reunion.
I shouted out WTF NO when he took that shot from the gunship.

This moment for me was changed as more crowning awesomeness appeared. The apex of awesomeness for me was:
"With all due respect, Admirals, I didn't represent one of your people, I represented one of mine."
First time I'd ever gotten the chills from a video game voice over.

Yup, one of the best lines in the entire game. A very polite way of saying F.U., Tali is with me, **** your petty bull****.

I also liked during Tali's trial, the Paragon way out. Basically anything Paragon where Shep stands up for someone is awesome/heartwarming in my books.
Crowning Moment of Funny for me is the high volus when doing the recruit Samara mission, specifically the Paragon interrupt. The other interrupts are usually big and dramtic ... this time ... *push*

^This. That entire scene was extremely well done.

As others mentioned, the 'everyone lives' ending/mission. Especially when the platforms are collapsing and Shepard saves someone (he saved Grunt on mine ... you'd think he'd be heavy, lol).

I liked the renegade option with Jack in the group. "Shep... that was just mean. Kinda funny though."

"Stupid human, you can't even hit what you aim at."
BANG!!!!!
I play as a Paragon, but man that felt good! I still get all sorts of amusmant out of that bit.
Another Tuchanka favorite ~ My FemShep bawling out and shaming the Scout into getting back to camp. I'm sure having the GuyShep doing this is amusing, but listening to FemShep rip him apart is quite fun.

Shepard says something along the lines of "After all you've been through Tali you deserve better than that," and she replies "I did get better, Shepard. I got you." I was playing femShep but I d'aww'd all the same.

Okay, being serious:
Samara shooting you down in the end. I actually felt sad. That's one of the incredibly few times a game has ever made me feel anything other than "****, yeah, did you see what I just did!?"
