I unlocked many acheivements where, I had used...a shotgun, throw, and barrier enough, i could have it for a subsequent character....
QUESTION: Those that mean I get those abilites if i do another playthrough? Will it overwrite my original game?

I'm not entirely sure what you're asking, but if you mean start a new charactver with a different class so you can unlock the skill specific achievements (like neural shock etc) then that new Shepard gets his/her own set of saves - all your different Shepards are kept separate, it won't have any affect on your previous Shep's saves
