Hi all,
I've just found Samara at Illium Spaceport, and she's been taken into custody. I've managed to get the elevator key off of the little guy, but as soon as I enter the elevator and get to the loading screen my game crashes.
Thought it might have been my point in the game, so I went off to do a few more recruitment missions and side missions, and this still occurs.
Any suggestions?

I am having the exact same issue, in the exact same spot.
Windows 7 Ultimate 64 bit
Athlon 64 X2 6000+
2 x Geforce 8800GTS
4 GB of RAM
Creative Sound Xtreme Gamer

I'm guessing that it could be a hardware issue. Ta, can you post your computer specs?

Also, I just tried taking my sound card out of my computer. The same result, you click on the elevator button and the loading screen pops up for about 3 seconds and then crash to the desktop. Sigh.

I suppose this could be something similar to the crash upon releasing Grunt some people have had. (That one seems to be video card-related.) But my money would be on a corrupt file - especially since no heavy rendering is involved here - as it would be with most recurring errors that are not widely experienced and occur well in to the game.
So: If you got the game through a Direct Download service like Steam, Impulse, etc. you may be able to tell the download client to "Verify" your installation and make sure all the files are intact. If you have the game on disc, or if your Direct Download service doesn't support verification for Mass Effect, you'll have to re-install.
When re-installing Mass Effect 2, make sure to tell the game to keep your settings & saves when you uninstall.
To try verifying your files on your Direct Download service:
In Steam, right-click on the game and select "Properties." Select the "Local Files" tab and click the button that says "Verify integrity of game cache..."
In Impulse, right-click on the game. On the menu that pops up, there may be an option to "Verify Installation." (I do not see this option for my Mass Effect 2 DDE. Can anyone comment on Mass Effect 1?)
Direct2Drive: Don't know, never used it. Can anyone comment?
EA Download Manager: Don't know, never used it. Can anyone comment?

I've already tried verifying my Steam install twice, and the same issue occurs.
