Like in Super Mario Galaxy, the game would still revolve around the first player, without the second detracting from the quality.

It already is. When multiplayer replayability is taken over plot, especially when it's competitive. Multiplayer doesn't HAVE to ruin story driven games depending on how it's handled.

Baldurs Gate is also based and uses D&D as a foundation. A RPG thats whole purpose is centered around group play. It is also not a shooter type of multiplayer which is likely was Mass Effect would de-evolve into if it became co-op.

What if co-op only engaged at the squad selection screen immediately before any missions? That is, Player 1 is playing as Shepard (hosting, so to speak) and Player 2 is a guest able to select whatever squaddie P1 has recruited.
They land the roach and shootshootshootie fun begins. Whenever there's a dialogue P2 can interact in a similar way to that planned for Star Wars: The Old Republic. Or, if you dont want P2 derailing your decisions then maybe just a bit of banter? (P2 as Grunt could get the option to growl and threaten an NPC without it affecting P1 Para/Ren score)
IF, and this is a monumental IF, this didn't affect the story telling (I honestly cant see how it could, provided it is handled carefully. And I trust Bioware in that regard) then I'm not sure if it'd be that bad.
Now that I think about it, reactions from P2 in my game as I shoot or talk to NPCs could add a lot of variety to the game. Now, I wouldn't go and invite random strangers to my game, of course. That woul be to invite disaster, same as with any other online mode.

Difference of opinion. I just shared mine and won't patronize anyone for sharing theirs. If you want it it's your choice. Personally I just wouldn't use the feature even if it was made available. I've played Multiplayer games, they're fun to an extent, but what has always appealed to me has been SINGLE player and Mass Effect pulls it off very well, so I just hope it stays that way. Plus in my honest opinion not every game out there needs multiplayer but everyone is hopping on that train before it leaves. Case in point Bioshock 2, did it really need multiplayer? So I just choose not to use it. I'll just do the same with ME. Assuming it does integrate a multiplayer co-op system.
And yeah it is possible to make a single player game with a co-op option to it, but it's very hard to do it right. Fable is one of the FEW games I can recollect that uses the system well enough to an extent. But even that has it's issues. But still I honestly don't speculate it working well enough with Mass Effect. As for the first point I'd assume the only reason anyone is asking serious consideration into a co-op mode for the game is because they want it to be an integral part of gameplay which might seem a little lackluster in it's current state due to the absence of co-op.

For some strange reason Bioware just doesnt want to admit their main sales comes from RPGs, not shooters and RPGers is what they should be focusing on.
Anyways, after reading Christina Normans hugely insulting power point presentation, Ive taken ME3 off my shopping list unless someone in the know comes out and say point blank this this and this regarding RPG elements has been fixed from ME2 version.
ME2 just wasnt fun as it should have been cause it just literally failed to live up to its original who set the bar so very high.
Best announcement I could hear is ME1 developers rehired or resigned to make up ME3 development team. Speaking for myself, I have no faith in the ME2 development team who lied to us and strung us along to get those extra sale numbers.

However, I can imagine a stand alone ME Co-op game which is developed from the ground up to feature that. But I am afraid that if you mix the two in one game then one of the two or probably both have to suffer. Also, BioWare has no expertise in that area. So, the first one will be a trial on error experience or the company has to outsource the project. The good doctors seem to have good contacts with DICE. Maybe they can dream up something cool together for the ME universe.

Personally, I think any focus on multiplayer comes at the expense of singleplayer. Look at Halo, CoD, Bad Company, etc. Their singleplayer is lackluster.
If you want to play with your friends, go play those. Don't invite EA to Firefly our Mass Effect.
