-How are they activated?
-Do people live in them/operate the jumps made by one?
-If no to ^, then how did they find out "send x signal to relay and you go to y"?(Prothean data archives perhaps?)
-Are they ever used as giant mass effect cannons?
-And last but not least, what powers them?

Not sure how they are exactly activated, probably just a matter of starting up its core. I don't believe they are manned at all, but they are connected to some kind of network. In ME1 the state that you can control the activation of any relay from the citadel, its how the Reapers usually wipe civilizations out, by turning off all the relays so they're isolated. I'm sure a computer in the relay sends all the trajectory data to the vessel using the relay to help chart their course. To date they've never been used as weapons, but if you closed a mass effect field on a ship in transit it would convert all it's matter into energy. The relays are all powered by Element Zero, the fictional element which is used for all FTL travel and responsible for manifesting biotic powers in exposed humans. When element zero is subjected to an electrical current it creates a field of dark energy which can raise or lower the mass of anything caught within it. Lowering the mass of a ship to near 0 makes FTL travel much easier and gets around the problem of creating infinite mass when exceeding the speed of light.

Every intelligent species in the universe has one common language - the language of science. So I don't think any species who has a basic understanding of maths and sciences will have any trouble figuring out the relays and activating them after a bit of research.
