Ryan Reynolds - If it's a male role, I pretty much always have to recommend Reynolds. Hell, even most female roles I recommend Reynolds. Also, he's incredibly fit looking without looking stupid buff. Though I'm not really sure if I'd want my Shepard to be funny. But he can do *anything.*
Nathan Fillion - Yeah, I know, I know, it's just because he was in Firefly. But I think he could strike a good balance between being funny and serious if the situation called for it.
Sharlto Copley - If you set up Shepard as being less of a soldier and more of a techie, or, preferably, a biotic, I would shove Copley in that role so fast that...uh...something would happen. Black hole, sure. I don't think he could pull off being a straight soldier, but I can totally see him slinging around some singularities.
Eric Bana - This guy would probably be my first choice for a soldier Shep. He's a bit older than the Hollywood norm (I think. He has some grey in his hair now, so it counts), so if they wanted to put emphasis on his career, he would be good for a "Shep with a past."

I would rather see them do what they did with Avatar and use the new technology to create an exact look like of the game characters. This was effectively done with the Navi in that movie so why couldn't it work with this. This way they have the people perform the action themselves and they could map the characters onto the actors. To me this seems to be the best route to go because we could have the original voices and the actual looks of the characters.
