I always skip all paragon and renegade options to get him to say that. It's so worth it.

1 - do you have to go through all of Mordin's other dialogue first to get the "advice", or will he automatically give it to you if you talk to him before the Omega relay, even if you've rarely been down to the lab to talk to him otherwise?
2 - Let's say you start a romance but then give them the "friends" talk *before* traveling to the relay (i.e. Shepard sits alone in the captain's quarters), will Mordin then accuse you of being interested in him (as opposed to the sex talk), or do you have to have *never* initiated romance dialogue with anyone in order to get that particular dialogue?
Hope those questions make sense...also since people post in here so infrequently maybe if you know the answer you could PM me so I don't miss it? Thanks!

Like one playthrough where I romanced Garrus, Mordin gave me the talk about me hitting on him. This is because I talked to Mordin constantly during that game, so I hit that part of his dialogue progression where the sex talk comes up before I hit the part of Garrus's dialogue progression where the romance officially starts.
Another playthrough, I romanced Thane and did get Mordin's sex talk about drell. This is because I had definitely established the romance with Thane before going through a lot of Mordin's dialogues.
So it doesn't matter really when you talk to Mordin or the love interest, but how often you do.

I wish I could remember the progression of Mordin's dialogue, but I believe the sex talk is always at the same point in the progression. So whether or not Mordin thinks you're hitting on him or gives advice on sex with another depends on where you are in the progression of the romance's dialogue.
This.
I got Mordin's 'Would try you' line /w my MaleShepard. At the time, I was planning on romancing Jack (before I switched to Tali), but I always spoke to Mordin first after every major assignment following his loyalty mission.

It was fine at first, but as the days drew on I felt I missed something from it. Something was calling me back to the game.
Andf then I realised it was none other then Mordin! I love that crazy little Salarian! I support Mordin for ME3.

Not sure if it's been mentioned, but it also apparently doesn't matter if Shepard is male, either.
I got the same "you seem interested, but I'm not" talk from him with my male Shepard, which struck me as odd, but amusing. Of course, I'd spent most of the game talking to Mordin more than anyone else save Joker simply because he almost always had something funny to say.
EDIT:
Yep, looks like someone else mentioned it as well.

Mordin is probably one of my favourite characters in a game ever. I won't be pleased if he's not in Mass Effect 3 as a squad member, because I made sure he lived. I gave up the chance to fight with him in the final mission, just because I knew sending him back to the ship as the escort was definitely going to keep him alive.
