With Mass Effect 1-3 conquering the 3rd person/rpg Combo what would you want 4-6 to cover.

Well, Mass Effect 3 will probably end with Shepard succombing to the Dark Side.
And 4-6 would be about Shepard's illegitimate son trying to bring down the evil Empire that his father worked so hard to establish.

That would be really nasty.
I think Bioware should go back to their roots and create a REAL Mass Effect RPG. I miss when you had to remember to drink water, and it took 5 hours to fly somewhere REAL TIME!

Space combat sim. More Wing Commander than Privateer/Elite. (Yes, I know the whole fighters-vs.-capital-ships thing is a little unrealistic, but I do not care, and fighters are mentioned in the Codex and novels but never shown.)

A Wing Commander type game would fit, as WC 3-5 had a major focus on story and character, something BioWare loves. Especially if you added RPG type elements into it (upgrades, skill acquisition, trees, etc) would make a unique space combat game.
Another idea, which would be killer in my book, would be an RPG/Adventure hybrid set entirely on the Citadel. Mood would be noir, with cheesy narration from the protaganist. It would be an opportunity to really open up the Citadel, each arm containing immense possibilities for story and gameplay. You would really get to see the seedy side of the ME universe. Plus, the main character would be a Garrus in a trench coat and fedora, playing detective, possibly butting heads with corrupt CSEC cops.
