Of course it changes the entire character. I can think of few things more character defining than one's sexual preference.
Once again I feel compelled to point out that, remember, these characters that we're talking about were originally bisexual, and that their characterizations and dialogues and involvement in the story were written around that basis. How would their potential bisexuality significantly deviate the story of ME2, considering that the story was originally written with it in mind?
