I just spent 2 hours playing Mass Effect 2, with the option to save constantly grayed out. Even after obtaining the Normandy, figuring I'd be able to save there, I'm still unable to save. What's the deal?

Woah, major breaking bug for you there. Ever tried restarting with a new character and see what's happening?
And I hope the Auto-save feature WORKS because the Auto-save feature saves at the exact moments you want to.
It saves when you visit vendors and buy stuff, at every location you [re]-enter, combat and whatnot.

No save whatsoever ... even though I saw the disk pop up and say auto saving, there is no auto save and unable to save on command ...
I just went through the hell of replaying all of Mass Effect 1 again because I was originally unable to import my character, and now this .... not happy

Try contacting EA support, which I contradictly not advice. Try contacting a Dev around here.

Is autosave turned on in your options menu? I'm guessing from what you said above that there is no autosave point listed in your load menu - ?

I've got the same problem. I bought ME about a month ago, finally completed it last night, and loaded up my copy of the ME2 Collector's Edition to import my save and start the new game. So far, I've tried twice, and haven't been able to save. I did what other people have, thought I had to get to the Normandy to be able to save, but no dice. At this point I just wasted the last 5 hours trying to get the freaking thing to save, to no avail. I certainly am not going to start a new character and forget about importing the one that I spent the last month leveling up, just so that I could import it. I certainly hope that Bioware figures this out and fixes it soon, because otherwise I will have a very expensive bookend sitting on my shelf that will never get played, and I certainly won't be buying ME3 if they're just going to release buggy crap.
In fact, I didn't have the autosave on, but there were points (when I was tweaking my character's outfit) where it said it was saving. I restarted the game, just to see if I could open those save files, and it's like they don't even exist.

Ok, so I got it to work, finally. The following is exactly what I did, in case it helps anyone else...
1. Disconnected the hard drive from my 360. Yes, this will shut it off, don't be surprised when it does.
2. Left it disconnected for about 1-2 minutes, then reconnected it and restarted my 360.
3. Loaded the game.
4. Started a new game (because it still was saying that there wasn't a previous save file).
5. Told it to import my ME character.
6. Did not try to modify my ME character in any way - just did a straight copy-over.
7. Told it to have the auto-save on (previously I had left it off).
8. Watched the beginning of the game for the 5th time.
9. As soon as I woke up in the med bay, it let me save, even before I got my pistol & armor.
I'm not sure which of the above steps worked, but it did, and it's been fine since. My fiance uses the same 360, and he did a straight copy-over with no modification of his save file, and his worked fine as well. I hope this helps!
