I haven't tried it, but I'm going to go ahead and say it's possible. At least it was in ME1.

Should technically be possible to install DLC, connect to internet, validate the DLC (automatic when connecting to Cerberus Network) and then disconnect forever.
Technically. The reality is very different and many people have had problems with it, resorting to third-party means. Ufortunately, this means that you can -never- connect again (it would invalidate your DLC) so you'll have to stand there, wondering why you even bothered to pay for it in the first place, and depend on the same third-party means of getting new DLC to work.
Edit: Oh, wait. Didn't see that this war for X360. Ignore the part about third-party means.

As long as it's the original console, then everything should be fine. I play offline regularly without any problem. However if you change console, then you need to transfer your gamertag licenses to be able to play dlc and xbla games offline.

I actually can't be 100% sure here, but if I'm not mistaken, I played ME1 offline at some point and managed to play through the DLC mission (BDts) with zero problems.
I think it's like Gad mentioned above; you just need to make sure that it's the same console/hard drive on which the DLC was originally downloaded. If all is well, then in theory, you should be able to play the DLC offline without the slightest issue. I've done it with other games, such as Rainbow Six: Vegas 2 (played the DLC maps offline), and even ME1 if my memory isn't betraying me.

Well... I downloaded all DLCs that were avaiable for me (Cerberus Armor & Shotgun, DrPepper Helmets, Zaeed, Normandy Crashsite) and then disconnected... I only connect If I want to check for Updates... and I can play without a problem...

On my xbox I have a few profiles, for my roommates and I. Recently my roommate has been unable to play because it says he doesn't have the crash site or zaeed files. I'm thinking it may have to do with the ea account being tied to my gamertag but I don't understand why it was working but now stopped. His account was never made into a gold account, would it work if he played with the ethernet cable out?
